Antibiotics have components to inhibit infections against Staphylococcus aureus, but they depend on judicious use to minimize the incidence of resistance forms. Strategies to improve the current situation include research in finding a new antimicrobial from virgin coconut oil (VCO). The saturated fatty acid, lauric acid (LA) (C12) contain in VCO was reported to have antibacterial activities. This study developed antimicrobial of VCO as an antimicrobial and immunomodulatory agent. Staphylococcus aureus used in this study had been isolated and identified from the mastitis milk crossbreed Etawa goat from Riau, Indonesia. The susceptibility of S. aureus to VCO was tested using the broth dilution method. The inhibition mechanisms of S. aureus had been studied by scanning electron microscopy (SEM) after treatment with VCO, and potential of VCO, which is known in phagocytosis macrophage. In vitro test confirmed the inhibitory effect of VCO on the growth of S. aureus at the concentration of 200 μl (equal to 0.102 % LA). Based on the result of the phagocytosing assay, VCO could increase the ability of the macrophage cells to phagocyte S. aureus significantly at a concentration of 200 μL (equal to 0.102% LA). This study concluded that the VCO could inhibit the growth of S. aureus with destructive mechanisms of bacterial cell walls and increase the ability of the phagocytic immune cells.
Aflatoxin contamination has been well known as a world-wide health-threatening problem in tropical countries including Indonesia. This research was undertaken to determine the degree of aflatoxin contamination in different Indonesian foodstuffs. A preliminary survey was carried out to evaluate the level of total aflatoxin (AfT) and aflatoxin B1 (AfB1) contamination of baby foods, peanut products, and corn products, which were purchased from traditional markets and supermarkets in Indonesia during the year 2001-2002. Eighty two peanut products, 12 baby foods products, and 11 corn products from different brands were analysed for AfT and AfB1 using the Enzyme-Linked Immunosorbent Assay (ELISA) method. The results indicate that, of the brands analysed, 35% of the peanut products were contaminated with aflatoxins at various levels (range 5 to 870 μg/kg). Peanut-chilli sauces had the highest percentage of AfT contamination 9/12 (75%), which was followed by traditional snacks 5/11 (45%), peanut butter 4/11 (40%), flour egg coated peanut 6/16 (37%), and peanut cake 3/10 (30%). Fried peanuts and roasted peanut were found to contain aflatoxin at relatively lower percentages of 9% and 8%, respectively. From the 12 analysed baby food samples, on the other hand, no sample was found to be contaminated with aflatoxins. Two of 11 samples (18%) of corn based products were contaminated with AfT, ranging between 5.8 and 12.4 μg/kg. Additionally, 30 selected samples in different concentration ranges were further analysed to verify the correlation between ELISA and HPLC techniques and results were compared.
A lactation trial was conducted to determine the effects of supplementing whole safflower seeds (SS) on ruminal fermentation, lactational performance, and milk fatty acid (FA) profiles. Nine multiparous Holstein cows (days in milk = 110 ± 20) were used in a replicated 3 × 3 Latin square design. Each period lasted 21 d, with 14 d of adaptation and 7 d of data collection. Within square, cows were randomly assigned to a sequence of 3 dietary treatments as follows: cottonseed total mixed ration (TMR; CST), conventional SS (variety S-208) TMR (CSST), and NutraSaff SS (Safflower Technologies International, Sidney, MT) TMR (NSST). Diets contained approximately 63% forage (36% alfalfa hay, 4% grass hay, and 23% corn silage) and 37% concentrate supplemented with 2% cottonseed to the CST and 3% conventional or NutraSaff SS to the CSST or the NSST, respectively. Intake of dry matter (DM) averaged 21.8 kg/d and did not differ across diets, but feeding the NSST decreased intake of neutral detergent fiber (NDF) due to lower dietary concentration of NDF in the NSST. Digestibilities of DM and nutrients were similar among treatments. No differences in yields of milk or milk components were observed in response to supplementing SS. Dietary treatments did not affect ruminal pH, total or molar proportions of ruminal volatile FA, and ammonia-N. However, cows fed SS had a higher molar proportion of isobutyrate than those fed the CST diet. Ruminal C16:0 FA concentration increased with the CST, whereas C18:1 cis-9 and C18:2 n-6 tended to increase with SS supplementation, indicating that conventional and NutraSaff SS were partially protected from microbial biohydrogenation. Supplementing SS decreased milk C16:0 concentration, whereas it increased C18:1 cis-9 and C18:1 trans-9. Milk FA C18:1 trans-11 and cis-9, trans-11 conjugated linoleic acid increased and tended to increase with feeding the NSST, respectively, but not the CSST diet. In conclusion, supplementing diets with whole SS at 3% of dietary DM can be an effective strategy of fat supplementation to lactating dairy cows without negative effects on lactational performance and milk FA profiles.
The objective of study was to estimate breeding value of sires of Kebumen Ongole grade cattle based on offsprings weaning weight. This research was carried out from August to December 2015 in Urut Sewu area, Kebumen. The calculation of genetic parameter estimation was heritability of weaning weight. The value of heritability was used to calculate the breeding value of sires based on offsprings weaning weight. Heritability was calculated using paternal half-sib correlation with the direct pattern variance analysis. Breeding Value (BV) of sire was calculated by absolute estimated breeding value. The materials used in this study included were data record during the last three years (2013-2015) consisted of 41 sires, 51 dams and 244 calves. Weaning weight data had been corrected based on age of dams, sex and weaning age of 120 days. The results showed that the heritability value of weaning weight was included in high category (0.36 ± 0.21). The highest BV was KJ.J608 (49.76) with a ratio of 231.45%. The results of this study were expected to be the basis recommendation for selecting sires in Kebumen.
