The disappearance of dry matter (DM), crude protein (CP), and amino acids (AA) in steers after rumen incubation and intestinal passage of alfalfa hay, barley hay, corn silage, barley grain, corn grain, wheat bran, meat meal, fish meal, cottonseed meal, and soybean meal were measured in 3 steers using a mobile nylon bag technique. Ruminal degradation of individual AA differed between feedstuffs. For barley hay and corn silage, the ruminal disappearance of total AA was higher and lower than the other feedstuffs, respectively. The intestinal digestibility of total AA in alfalfa hay was lower than the digestion of CP. The intestinal digestibility of Arg and His was higher than that of total AA in alfalfa hay, meat meal, cottonseed meal, soybean meal, barley hay, and wheat bran. In addition, the intestinal digestibility of Lys was higher than that of total AA in alfalfa hay, meat meal, cottonseed meal, soybean meal, barley hay, corn silage, and wheat bran. The intestinal disappearance of CP in most cases was higher than that of DM. The results indicated that feedstuffs with lower ruminal disappearance of DM, CP, total AA, essential AA, and nonessential AA generally had a higher intestinal disappearance, resulting in a relatively constant total tract disappearance. These results could be used to improve the current system of diet formulation in ruminants.
The bovine lymphocyte antigen (BoLA-DRB3) gene encodes cell surface glycoproteins that initiate immune response by presenting processed antigenic peptides to CD4 T helper cells. DRB3 is the most polymorphic bovine MHC class II gene which encodes the peptide-binding groove. DRB3 gene has been extensively evaluated as a candidate marker for association with various bovine diseases and immunological traits. This study describes genetic variability in the BoLA-DRB3 in Iranian Holstein cattle. This is the first study of the DNA polymorphism of the BoLA-DRB3 gene in Iranian Holstein cattle. Hemi-nested PCR-RFLP method is used for identification the frequency of BoLA-DRB3 alleles. The BoLA-DRB3 locus is highly polymorphic in the studied herd (26 alleles). Almost 67% of the alleles were accounted for four alleles *24,*11,and *16 ) in Iranian Holstein cattle. The DRB3.2*8 allele frequency (26.6%) was higher than the others. The frequencies of the DRB3.2*54, *37, *36, *28, *25, *14, *13, *10, *1 alleles were lower than 1%. Significant distinctions have been found between Iranian Holstein cattle and other cattle breeds studied. In Iranian Holstein cattle the alleles ( BoLA-DRB3.2*22, *2, and *16 ) associated with a lower risk of cystic ovarian disease in Holstein cattle are found. The alleles associated with the resistance to mastitis and to bovine leukemia virus infection BoLA-DRB3.2*11 and *23 are detected with the frequencies 10.4 and 4.4%, respectively. Thus, in the Iranian Holstein cows studied alleles associated with resistance to various diseases are found. The method of DNA-typing of animals can be used in agricultural practice for BoLA-DRB3 allele genotyping of cattle in order to reduce spreading of alleles providing susceptibility to mastitis or leukemia in cattle herds.
Chemical composition and physical characteristics of diet influences its optimal utilization in dairy cow. Neutral detergent fiber measures the chemical characteristics, but not the physical characteristics of fiber such as particle size and density (Mertens, 1997). Both the amount and physical form of fiber are important in the diet of lactating dairy cow for maintaining normal milk fat content, chewing and digestive activities. A sufficient supply of long particles or NDF must be included in the ration to increase chewing activities, maintaining the rumen pH and providing an optimal rumen environment in order to avoid metabolic disorders. Current NRC guidelines (NRC, 2001) have proven useful information in defining animal requirements and feed composition but do not provide detailed recommendation on physical form of ration. The objective of this experiment was to determine the effect of lucerne particle size on feed intake and digestibility in dairy cow in early lactation period.
