Automated production of a promising radiopharmaceutical (-)-(1-(8-(2-[18F]fluoroethoxy)-3-hydroxy-1,2,3,4-tetrahydronaphthalen-2-yl)-piperidin-4-yl)(4-fluorophenyl)methanone ([18F]VAT) for vesicular acetylcholine transporter(VAChT) was achieved using a two-step procedure in a current good manufacturing practices fashion. The production of [18F]VAT was accomplished in approximately 140 min, with radiochemical yield of ~15.0% (decay corrected), specific activity > 111 GBq/μmol, radiochemical purity > 99% and mass of VAT ~3.4 μg/batch (n > 10). The radiopharmaceutical product meets all quality control criteria for human use, and is suitable for clinical PET studies of VAChT.
Washington University has produced radioisotopes for medical application since the early 1960s. In order to serve seven PET scanners and to meet more stringent government regulations we have installed a new PET pharmacy based on our past years of experiences. The new pharmacy was installed at the site of the 3.7 MeV tandem cascade accelerator that was decommissioned in April of 2001. The pharmacy consists of a production lab, quality control lab, reagent preparation lab, shipping and storage area and an office. Security and safety was a main consideration in the design of this PET pharmacy. A self shielded CTI RDS111 cyclotron was installed to supply the following precursors, 18 F-Fluoride, 18 F-fluorine, 11 C-carbon dioxide, 11 Ccarbon monoxide, 13 N-ammonia, 15 O-oxygen, 15 O-carbon dioxide and 15 O-carbon monoxide. 15 O-oxygen, carbon monoxide and 15 O-water
