Heat shock proteins (HSP) are a kind of chaperones, up-regulated after various stress reactions. HSPs classified on molecular sizes, HSP 90 inhibitor have been tried a adjuvant therapy on the treatment of melanoma. We evaluated apoptosis and necrosis of human normal melanocyte and melanoma cell line treated with HSP90 inhibitor, 17-Dimethylaminoethylamino-17-demethoxygeldanamycin, 17-DMAG), under hyperthermia condition. SK-MEL-2 cells were stimulated at 37 C or 43 C for 1 h and treated with 0M, 100nM and 1mM of 17-DMAG. 17-DMAG treatment increased apoptotic populations about 2.17% (100nM) and 3.05% (1mM) in 37 C controls and about 4.40% (100nM) and 4.97% (1 mM) in 43 C stimulated groups, and necrotic populations about 0.15% (100nM) and 0.66% (1mM) in 37 C controls and about 0.04% (100nM) and 1.62% (1 mM) in 43 C stimulated groups.
