Background and Aim: Reproductive traits play an important role in population increases and the egg production (EP) abilities of Indonesian local ducks (ILD). The prolactin (PRL) gene is a single chain polypeptide hormone belonging to a family of growth hormone genes that are mainly synthesized in the anterior pituitary gland in all vertebrates. It has a significant effect on reproductive traits and EP. Single nucleotide polymorphisms (SNPs) present in PRL are a useful molecular marker for EP. This study aimed to identify the PRL polymorphisms based on these SNPs and to uncover the associations with reproductive traits in ILD. Materials and Methods: A total of 280 ILDs consisting of Tegal and Magelang (F0) ducks and their reciprocal crosses, namely, Gallang (F1) and Maggal (F1), were maintained and specific variables were recorded, that is, age at first egg, body weight at first egg, first egg weight, and EP, for 90 days. Allele and genotype frequencies were used to determine the Hardy- Weinberg (H-W) equilibrium. The association between the SNP genotypes of PRL and reproductive traits was analyzed using one-way analysis of variance, following the GLM procedure of SAS. The genotypic effects on the reproductive traits were determined using regression analysis. Results: This study successfully amplified a polymerase chain reaction product of 190 bp, which was used to identify the SNP. Results indicated that PRL in ILDs is polymorphic. A SNP was found at position 164 nt (c.164G >A), consisting of three different genotypes, namely, GG, GA, and AA. The genotypes of Tegal and Magelang (F0), and Gallang (F1) populations were not in H-W equilibrium. The Maggal population (F1) was in H-W equilibrium. Significant associations were detected between the genotypes and EP in all ILDs (p<0.01), following a regression line of y=2.337x+64.605, with a determination coefficient of 0.0188 (r=0.14). Conclusion: PRL can be recommended as a candidate gene for reproductive traits in ILD, especially EP.
Aim: The present study was to investigate the interaction between duck's breed and phytogenic compounds as feed additives in the diet on blood lipid and hematological profile, welfare, and growth performance. Materials and Methods: A total of 200 male day-old local breed ducks (Tegal and Muscovy ducks) were used in this experiment. The first factor was duck breed and the second factor was different phytogenic compounds supplementation in the diet: Garlic, turmeric, ginger, and kencur, at 3% each. The observed variables were the blood lipid profiles comprise high-density lipoprotein (HDL), low-density lipoprotein, cholesterol total, triglyceride, blood parameters, welfare (heterophil/ lymphocyte [H/L] ratio), and growth performances (feed consumption, body weight gain, feed conversion ratio, and carcass percentage). Results: The interaction between breed of ducks and phytogenic compounds had a significant effect on blood triglyceride, but no significant effect on the blood lipid profile, hematological parameters, and growth performances. While, phytogenic compounds in the diet had significant effects on the blood lipid profile, heterophil (H), lymphocyte (L), and H/L ratio of ducks. The breed factors affected HDL and growth performances. Muscovy duck had a higher HDL and growth performance compare to Tegal duck. Among those, garlic most effectively reduced triglyceride level in Tegal duck. Conclusion: Phytogenic compounds 3% do not have a negative effect on the physiological parameters of ducks increase ducks welfare (H/L ratio), so it does not affect the growth performances of ducks. Muscovy duck had higher growth performances than Tegal ducks.
The aims of this research was to identify the qualitative and quantitative difference of the phenotypes of native mallard and Muscovy, and Muscovy the genetic diversity between Muscovy and mallard using RFLP method with growth hormone primer (GH) gene. The materials were 30-week old male and female ducks from 5 strains—Magelang, Mojosari and Tegal ducks, white-feathered, and black and white-feathered Muscovy. Muscovy Completely Randomized Design was used to analyze parameters 5 strains of ducks based on gender and with 5 replications. The parameters measured were local and Muscovy duck phenotypes and genetic diversity (number of alleles) based on GH gene, heterozygosity and genetic distance. Qualitative properties were analyzed descriptively and quantitative properties were analyzed using Anova followed by HSD in case of significant differences, whereas RFLP analysis was used to determine the allele frequencies, genotype frequencies, genetic diversity, and genetic distance of local ducks. Result showed phenotypic differences between Anas platyrhynchos and Cairina moschata. The dominant feather colorof Muscovy was black and white while mallard was brown. The bill color Muscovy Muscovy was pink with dark brown but black in mallard. The dominant shank’s color in both Muscovy and mallard was black. Body size of mallard was larger than that of Muscovy Muscovy, and Magelang ducks weigherd more than Tegal and Mojosari Duck. The PCR-RFLP results showed lower heterozygosity of mallard compared to Muscovy based on GH gene, and genetic distance of the Tegal duck was closer to Mojosari and Magelang ducks, whereas Magelang and Mojosari ducks had a considerable genetic distance based on the GH gene.
Abstract. Phytobiotic properties of garlic (Allium sativum), turmeric (Curcuma domestica), red ginger (Zingiber officinale) and kencur (Kaempferia galangal) were studied using standard in vitro antibacterial test and in vivo feeding trial with ducklings. In the in vitro experiment, potency of aqueous extract of these phytobiotic agents were tested against Salmonella pullorum and Escherichia coli. Feeding trial was carried out for 6 week starting at day 28 using ducklings fed diets supplemented with 1% of each of four phytobiotic agents. The highest antibacterial activity against S. pullorum and E. coli was observed with garlic and no additive effect when mixture of phytobiotics was used. Weight gain, fed intake and feed conversion ratio of ducklings were not affected by inclusion of garlic, red ginger and kencur. However, 1% turmeric supplementation significantly reduced growth performance to ducklings.Key words: phytobiotic, antibiotic, duck, medicinal plants Abstrak. Penelitian karakteristik fitobiotik dari bawang putih (Allium sativum), kunyit (Curcuma domestica), jahe merah (Zingiber officinale) dan kencur (Kaempferia galangal) telah dilakukan secara in vitro melalui uji aktivitas antibakteri dan secara in vivo dengan perlakuan suplemetasi fitobiotik didalam pakan anak itik. Pada percobaan in vitro, potensi aktivitas antibakteri dari ektrak fitobiotik diuji menggunakan Salmonella pullorum and Escherichia coli. Percobaan suplementasi fitobiotik diberikan masing-masing sebesar 1% didalam pakan anak itik. Pemberian pakan perlakuan dilakukan selama 6 minggu, dimulai pada saat anak itik berumur 28 hari. Hasil penelitian menunjukkan aktivitas antibakteri terhadap S. pullorum dan E. coli paling tinggi adalah ektrak bawang putih dan tidak ada pengaruh yang lebih baik apabila dicampur dengan ektrak fitobiotik lainnya. Pertambahan bobot badan, konsumsi pakan dan konversi pakan anak itik tidak dipengaruhi oleh penambahan bawang putih, kunyit, jahe merah dan kencur. Akan tetapi, suplementasi kunyit nyata menurunkan performan pertumbuhan anak itik.
This study was conducted to compare the environmental conditions around farms which was located in Tegal for Tegal ducks and Magelang for Magelang ducks. This study was also aimed to compare the composition of both feed nutrient content and the egg production. The research was conducted by survey method with purposive random sampling based on duck populations and age (8-15 months) provided by the farmer. Each area was provided 10 farmers for every farm and the data were collected in July-September, 2017. Environmental condition measurement were the temperature of the duck housing, and the composition and nutrient content of feed and the egg production of ducks. The results showed that all farmers’ ducks on a dry system rearing (without water pool) with average temperature in Tegal area 30.53±1.38oC and Magelang 28.32±1.00oC. Feed compositions given for Tegal duck consist of: dried rice, rice bran, concentrate, trash fish, golden snail, vermicelli waste and shrimp waste. The average nutrient content was crude protein 19.25%, energy 2,913 kcal/kg, crude fiber 5.82%, crude fat 6.87%, Ca 3.06% and P 1.40%. Feed composition for Magelang ducks consisted of rice (nasi aking), rice bran and concentrate, with crude protein content of 17.99%, 2,801 kcal/kg, crude fiber 8.14%, crude fat 6.10%, Ca 2.29% and P 1.04%. Tegal duck egg production was lower than Magelang ducks (64.89 vs 75.44%), but the egg weight was relatively the same. It could be concluded that the temperature of Tegal duck housing in Tegal region was hotter than Magelang region. Although the food quality of Tegal ducks was better than that of Magelang ducks, Tegal duck had a lower egg production because of the high environmental temperature.
