A simple and rapid method was developed for quantitative determination of common tetracyclines, such as oxytetracycline (OTC), tetracycline (TC), and chlortetracycline (CTC), in meat and fish. Tetracyclines were extracted with aqueous HCI, and then centrifuged. The supernate was applied to an Amberlite XAD-2 column, which was washed with water and eluted with methanol. The eluate was concentrated to about 0.5 mL under vacuum at 35°C, and then measured by liquid chromatography and UV detection. Two analytical columns were used for confirmation. The average recoveries of OTC, TC, and CTC from meat and fish fortified at 1, 1, and 3 ppm were 82.6, 81.5, and 67.0% respectively.
A simple, rapid, efficient method has been developed for determining thiabendazole, o-phenylphenol, and diphenyl in citrus fruits by using high performance liquid chromatography, with fluorescence or ultraviolet detection. The compounds are extracted with ethyl acetate and separated from soluble fruit components on a LiChrosorb RP-8 column. Recovery of these compounds added to citrus fruits at 5 or 50 ppm levels was >93%; the limit of detection for the compounds is 1 ppm.
An ion-pair high performance liquid chromatographic (HPLC) technique is described for the determination of inosinic acid (IMP) in meat. The compound was extracted with perchloric acid and analyzed without cleanup. IMP is effectively separated, identified, and quantitated by using a reverse phase column, with ultraviolet detection. A C8 stationary phase and tetrabutyl ammonium as counter ion are used. Recovery of IMP added to meat at 500 or 2500 ppm levels was more than 95%; the limit of detection for IMP is 50 ppm.
