ABSTRAK Pengobatan kanker menggunakan bahan alam terus dikembangkan. Salah satu tanaman yang memiliki efek sitotoksik Ocimum basilicum, L. tujuan penelitian ini adalah mengetahui aktivitas sitotoksik dari ekstrak etanol Ocimum basilicum (EEOB) terhadap sel kanker payudara T47D dan MCF7. Ocimum basilicum diekstraksi menggunakan metode maserasi dengan pelarut etanol 70%. Pengujian aktivitas sitotoksik menggunakan metode MTT assay dengan seri konsentrasi EEOB 1000; 500; 250; 125; 62.5; 31,25 µg/mL. Hasil uji aktivitas sitotoksik EEOB memperlihatkan nilai IC50 pada sel T47D dan MCF-7 sebesar 399.86 µg/ml dan 387.76 µg/ml. One of the plants that is proven to have cytotoxic activity is basil leaves (Ocimum basilicum, L.). This study aims to determine the cytotoxic activity of ethanol extract of basil leaves (EEBL) on T47D and MCF-7 breast cancer cells. Basil leaves were extracted using maceration with ethanol 70%. The cytotoxic test was perfomed using MTT assay with various EEBL concentrations: 1000; 500; 250; 125; 62.5; 31,25 µg/mL. Daun sambiloto dan kayu manis mengandung banyak komponen kimia yang dapat digunakan sebagai agen immunostimulan diantaranya andrographolid pada sambiloto dan sinamaldehid pada kayu manis. Penelitian ini bertujuan untuk mengkaji efek immunostimulan dari kombinasi eksrak etanol sambiloto dan kayu manis dengan melihat ekspresi sistem imun humoral (IgM dan IgG) pada mencit balb/c yang telah di induksi vaksin hepatitis B. Ekstraksi kedua bahan tersebut menggunakan etanol 96% dengan cara soxhletasi kemudian dikentalkan dengan rotary evaporator. Pengamatan sistem imun Humoral IgG dan IgM dengan menggunakan ELISA tidak langsung. Perlakuan pada hewan uji dilakukan dengan pemberian oral pada masing-masing ekstrak 150 mg/kgBB dan kombinasi ekstrak sambiloto dan kayu manis 1:1 , 1:2, dan 2:1. Data ELISA yang diperoleh adalah OD (Optical Density) yang menunjukkan jumlah imunoglobulin (IgG dan IgM) dalam serum. Hasil penelitian menunjukkan bahwa kombinasi ekstrak sambiloto dan kayu manis dengan dosis 300 mg : 150 mg menunjukkan peningkatan IgG sedangkan kadar IgM tertinggi pada kelompok ES:EKM 1:2 (dosis 150 mg : 300 mg). Ocimum basilicum L has proven to have in vitro cytotoxic activity against breast cancer cells. Pathways that cause cell death can involve one of the proteins, which is BCL-2. This study aims to determine the decrease of BCL-2 protein expressions in breast cancer cells (T47D and MCF-7) tat are treated with the ethanol extract of Ocimum basilicum L. Ocimum basilicum L. was extracted using the maceration method with 70% ethanol solvent. The concentration of ethanol extract of Ocimum basilicum L. used to see the expression of BCL-2 protein in T47D and MCF-7 cells was 199 µg/ml and 388 µg / mL. The observation of BCL-2 protein expression is using immunocytochemical methods of T47D and MCF-7 cancer cells. The results showed that the ethanol extract of Ocimum basilicum L could reduce BCL-2 protein expression in breast cancer cells (T47D and MCF-7) at concentrations of 199 µg/ml and 388 µg/ml, respectively.
