Despite the economical importance of sugar cane, until the present-date no studies have been carried out to determine the correlation of the molecular-based genetic similarity (GS) and the coefficient of parentage ( f)-estimates generated for cultivars. A comprehensive knowledge of the amount of genetic diversity in parental cultivars, could improve the effectiveness of breeding programmes. In this study, amplified fragment length polymorphism (AFLP) and pedigree data were used to investigate the genetic relationship in a group of 79 cultivars (interspecific hybrids), used as parents in one of the Brazilian breeding programmes, and four species of Saccharum ( Saccharum sinense, Saccharum barberi and two of Saccharum officinarum). The objectives of this study were to assess the level of genetic similarity among the sugar-cane cultivars and to investigate the correlation between the AFLP-based GS and f, based on pedigree information. Twenty one primer combinations were used to obtain the AFLP molecular markers, generating a total of 2,331 bands, of which 1,121 were polymorphic, with a polymorphism rate, on average, of 50% per primer combination. GSs were determined using Jaccard's similarity coefficient, and a final dendrogram was constructed using an unweighted pair-group method using arithmetic average (UPGMA). AFLP-based GS ranged from 0.28 to 0.89, with a mean of 0.47, whereas f ranged from 0 to 0.503, with a mean of 0.057. Cluster analysis using GS divided the genotypes into related subgroups suggesting that there is important genetic relationship among the cultivars. AFLP-based GS and f were significantly correlated ( r= 0.42, P< 0.001), thus the significance of this r value suggests that the AFLP data may help to more-accurately quantify the degree of relationship among sugar-cane cultivars.
Background Sodium-glucose cotransporter 2 inhibitors (SGLT2i) improve endothelial dysfunction and reduce cardiovascular events in individuals with type 2 diabetes (T2D). Proprotein convertase subtilisin/kexin 9 (PCSK9i) inhibitors reduce cardiovascular events in high-risk patients. Whether the addition of PCSK9i to SGLT2i treatment adds benefits is not known. Objectives To assess the PCSK9-i effect on the endothelial function of T2D individuals under treatment with SGLT2-i. Methods Individuals with T2D were randomized in a 1:1 ratio to a 16-week treatment with either empagliflozin (E) or empagliflozin plus evolocumab (EE). The primary endpoint was post-treatment change from baseline in flow-mediated dilation (FMD) at 1-min. Secondary outcomes included changes in plasma levels of nitric oxide metabolites and isoprostane. Results A total of 110 patients were enrolled, the mean age was 58 years, and 71% were men. The median post-treatment change in FMD at 1-min was 2.7% (interquartile range [IQR]: 0.9%) and 0.4% (IQR: 0.9%) in the EE and E groups, respectively (p < 0.001). There was a greater increase in plasma levels of nitrate [5.9 (16.5) vs. 2.6 (11.8); p = 0.001] and nitrite [0.14 (0.72) vs. 0.02 (0.74); p = 0.025] in the EE group than in the E group, respectively. Isoprostane reduction was more pronounced in the EE group when compared to the E group [−1.7 (5.9) vs. −1.1 (5.3); p < 0.001). Conclusions In individuals with T2D, the addition of evolocumab on top of empagliflozin improves endothelial function.
Background.Optimal control of traditional risk factors only partially attenuates the exceeding cardiovascular mortality of individuals with diabetes. Employment of machine learning (ML) techniques aimed at identi cation of novel features of risk prediction is a compelling target to tackle residual cardiovascular risk.Objective.To identify clinical phenotypes of T2D which are more prone for developing cardiovascular disease. Methods.The Brazilian Diabetes Study is a single-center, ongoing, prospective registry of T2D individuals. Eligible patients are 30 years-old or older, with a con rmed T2D diagnosis. After an initial visit for signature of informed consent form and medical history registration, all volunteers undergo biochemical analysis, echocardiography, carotid ultrasound, ophthalmologist visit, dual x-ray absorptiometry, coronary artery calcium score, polyneuropathy assessment, advanced glycation end-products reader, and ambulatory blood pressure monitoring. A 5-year follow-up will be conducted by yearly phone interviews for endpoints disclosure. The primary endpoint is the difference between ML-based clinical phenotypes in the incidence of a composite of death, myocardial infarction, revascularization, and stroke. Since June/2016, 1030 patients (mean age: 57 years, diabetes duration of 9.7 years, 58% male) were enrolled in our study. Mean follow-up time was 3.7 years in October/2021. Conclusions.The BDS will be the rst large population-based cohort dedicated to the identi cation of clinical phenotypes of T2D at higher risk of cardiovascular events. Data derived from this study will provide valuable information on risk estimation and prevention of cardiovascular and other diabetes-related events.ClinicalTrials.gov Identi er: NCT04949152
ABSTRACT.Isolates of entomopathogenic fungus Metarhizium anisopliae var. anisopliae were characterized using internal transcribed spacer-RFLP, ISSR and intron splice site primers. Thirtyseven isolates were studied, most of which were obtained from the sugar cane pest, Mahanarva fimbriolata (Hemiptera: Cercopidae) from Tangará da Serra, Southwest Mato Grosso State, Brazil. Internal transcribed spacer-RFLP did not differentiate the isolates of M. anisopliae var. anisopliae, while ISSR and intron primers identified three distinct groups. Variability among these groups was 96% for (GTG) 5 and 100% for the other primers. We found considerable genetic variability, even among isolates from the same geographical origin and host.
Este trabalho apresenta relatos e reflexões nascidas no período de Estágio Supervisionado do Curso de Licenciatura em Computação, do Núcleo de Ensino Superior de Coari (NESCOA) da Universidade do Estado do Amazonas (UEA), focando nas vivências junto a turmas de Ensino Médio da Escola Estadual Maria Almeida do Nascimento. A metodologia é qualitativa, composta por três etapas: observação, coparticipação e regência de classe. Foi diagnosticado que os docentes não utilizam ferramentas digitais em sua prática pedagógica ainda que a escola disponha de dispositivos tablet para os alunos. A utilização de recursos digitais foi negociada com os professores e partiu da realidade do contexto escolar. Por fim, a necessidade de formação permanente é problematizada.
