Tooth extraction is a common treatment in dental practice. However, complications such as bleeding or dry socket may occur during healing process. Pangas catfish (Pangasius djambal) gelatin contains glutamine which could increase the proliferation of fibroblast, accordingly collagen synthesis increases. The study aimed to demonstrate collagen density after the application of pangas catfish gelatin on post-tooth extraction wound of albino rats (Rattus norvegicus). This wasan experimental study with randomized posttest-only control group design which involved male Wistar strain albino rats. The rats were selected using a random sampling method and divided into four groups, namely day-3 control group (n=7),day-7 control group (n=7), day-3 treatment group (n=7) and day-7 treatment group (n=7). Pangas catfish gelatin was administered into the socket once after extraction. The variable measured in the study was collagen density, determinedby collagen scoring of hematoxylin eosin–stained histopathological preparations. Collagen formation were determined according to the assessment criteria of collagen distribution and density. Data analysis showed there was a significantdifference of collagen synthesis between the control and treatment groups according to Kruskal-Wallis test of which the result of control group day-3 was 1.13 ± 0.34, control group day-7 was 1.76 ± 0.67, treatment group day-3 was 1.66 ±0.47 and treatment group day-7 was 2.93 ± 0.25. In addition, the day-7 treatment group showed the highest collagen scoring values as compared to the control groups according to the Mann-Whitney test (p<0.05). In conclusion, pangascatfish gelatin increased collagen density during dental socket healing process after tooth extraction in albino rats.
Periodontitis merupakan keradangan pada jaringan pendukung gigi yang menyebabkan kerusakan tulang alveolar. Bakteri Aggregatibacter actinomycetecomitans menghasilkan Lipopolisakarida (LPS) yang dapat meningkatkan aktivitas osteoklas. Teh hijau memiliki kandungan epigallocatechin gallate (EGCG) yang dapat menurunkan aktivitas dan diferensiasi sel osteoklas dengan menghambat peningkatan RANKL. Tujuan: untuk mengetahui efek ekstrak daun teh hijau (Camellia sinensis) terhadap penurunan jumlah sel osteoklas tulang alveolar tikus putih (Rattus norvegicus) dengan diinduksi lipopolisakarida. Metode: Tikus putih sebanyak 28 ekor dibagi dalam 4 kelompok, yaitu Kelompok K-(tidak diberi perlakuan), Kelompok K+ (diinduksi LPS Aa), Kelompok P1 dan Kelompok P2 (masing-masing diinduksi LPS Aa dan diberi ekstrak daun teh hijau dengan dosis 150 mg / 100 gram BB dan 200 mg / 100 gram BB). Tikus didekaputasi untuk diambil tulang alveolar rahang bawah dan dilakukan pengamatan sel osteoklas dengan pewarnaan HE. Hasil: terdapat perbedaan jumlah sel osteoklas yang signifikan antar kelompok (One Way Anova, p<0,05) dan tidak terdapat perbedaan jumlah sel osteoklas yang signifikan antar kelompok P1 dan P2 (Post Hoc, p>0,05). Kesimpulan: ekstrak daun teh hijau (Camellia sinensis) mempunyai efek terhadap penurunan jumlah sel osteoklas tulang alveolar tikus putih (Rattus norvegicus) dengan diinduksi lipopolisakarida dimana dosis efektif ekstrak daun teh hijau 150mg/100grBB.
Abstract-Tooth extraction is always followed by wound healing process. Macrophage plays role as a phagocytosis cells, inducing granulation tissue formation, reepitelisation and angiogenesis process. Avocado (Persea americana) is a tropical fruits due to flavonoids contained has many benefits like provide immunostimulatory effects so that it will increase macrophage activation. The purpose of this research was determined the effect of avocado ethanolic extract for increasing macrophages cells after tooth extraction in Rattus norvegicus. This research was using randomized post-test only control group design method. Twenty four rats had tooth extracted. They were divided into 4 groups (K, P1, P2 and P3). In group K, subjects were not given avocado extract (control positive groups), group P1 were given 150mg/weight/day, group P2 300mg/weight/day and groups IV 450mg/weight/day orally for 3 and 7 days. The number of macrophages in rat's socket was determined by histological preparations with HE staining using a light microscope. As a result, the statistical test in day-3 showed differences in the number of macrophages cells between group (ANOVA, p = 0,000) and positive correlation between the dose of the avocado extract with macrophage cells number (r = 0,684). In day-7 showed differences between groups (Krusskal-Wallis, p = 0,969) but negative correlation between the dose of the avocado extract with the number of macrophages cells (r = -0,043). Conclusion : The avocado extract increased macrophage cells in rat's socket on day-3 in all doses but decreased on day-7 after tooth extraction.
Latar belakang: Tanaman herbal telah banyak digunakan masyarakat sebagai obat untuk menyembuhkan luka dengan efek samping yang minimal. Salah satu tanaman yang memiliki banyak manfaat, yaitu kulit jeruk nipis (Citrus aurantifolia Swingle) yang diketahui memiliki kadar flavonoid tinggi, sehingga dapat mempercepat proses penyembuhan luka dari ulkus. Pada fase penyembuhan luka, makrofag mempunyai peran utama sebagai fagosit dan sekresi growth factor. Untuk memaksimalkan efektivitas penghantaran obat, ekstrak kulit jeruk nipis dibentuk dalam sediaan nanoTransfersom sistem. Tujuan: untuk mengetahui pengaruh pemberian nanotransfersom ekstrak kulit jeruk nipis (Citrus aurantifolia Swingle) terhadap jumlah makrofag pada penyembuhan luka mukosa labial tikus. Metode: Penelitian ini menggunakan true experimental in vivo dengan induksi termal mukosa labial tikus Wistar, terbagi 4 kelompok dengan time series hari ke-3 dan 7, yaitu kontrol negatif [K(-)], kontrol positif diberi triamcinolone acetonida 0,1% [(K(+)3], perlakuan yang diberi ekstrak kulit jeruk nipis (PA), dan diberi nanoTransfersom ekstrak kulit jeruk nipis (PB). Jumlah makrofag diamati menggunakan mikroskop perbesaran 400x. Hasil: Analisa data menggunakan uji One Way Anova menunjukkan terdapat perbedaan yang signifikan (p<0.05) pada jumlah makrofag antar kelompok pada hari ke-3 dan ke-7. Kesimpulan: NanoTransfersom ekstrak kulit jeruk nipis (Citrus aurantifolia Swingle) berpengaruh terhadap jumlah makrofag pada penyembuhan luka mukosa labial tikus wistar.
