SUMMARY The human TEAD family of transcription factors (TEAD1-4) is required for YAP-mediated transcription in the Hippo pathway. Hyperactivation of TEAD’s co-activator YAP contributes to tissue overgrowth and human cancers, suggesting that pharmacological interference of TEAD-YAP activity may be an effective strategy for anticancer therapy. Here we report the discovery of a central pocket in the YAP-binding domain (YBD) of TEAD that is targetable by small molecule inhibitors. Our X-ray crystallography studies reveal that flufenamic acid, a non-steroidal anti-inflammatory drug (NSAID), binds to the central pocket of TEAD2 YBD. Our biochemical and functional analyses further demonstrate that binding of NSAIDs to TEAD inhibits TEAD-YAP-dependent transcription, cell migration and proliferation, indicating that the central pocket is important for TEAD function. Therefore, our studies discover a novel way of targeting TEAD transcription factors and set the stage for therapeutic development of specific TEAD-YAP inhibitors against human cancers.
This study aims to analyze the al-Qur'an reception at Al-Husna Islamic Boarding School Sidorejo Pamotan Rembang. This research is a qualitative research using inductive descriptive method. Qualitative research is a research procedure that analyzes descriptive data in the form of speech or written observations. Inductive descriptive method aims to describe and analyze data to obtain a general description or description of the al-Qu'ran reception at Al-Husna Islamic Boarding School. This research confirms that the Qur'an is a holy book that is always in harmony with all situations and conditions (ṣāliḥ li kulli zamān wa makān). The theory used by the researcher here is Karl Mannheim's sociological theory of meaning, which focuses on three typologies of meaning, namely objective, expressive, and documentary meanings. This study yielded at least two findings. First, there were several types of receptions at the Al-Husna Islamic Boarding School. Second, the meanings inherent in the reception include, a) objective meaning, that the variety of reception behavior at Al-Husna Islamic Boarding School is part of a symbol of obedience and disbelief towards the pondok regulations, b) the meaning of expression is manifested in the form of self-internalization with values positive values in the continuous learning process of the Koran, and c) documentary meaning is manifested in the form of local contextualization of a comprehensive cultural system.
In the Industrial Job Training system that is currently running in SMK, there are still obstacles such as the registration process, filling out journal books and the student assessment process for Industrial Work Practices. With a web-based industrial work practice information system can provide information about industrial work practice programs online. This system has advantages in terms of speed of presentation of the resulting information, in addition to being web-based, this system can be accessed at an unspecified time and place. The purpose of this study is to design an administrative information system that can provide information related to the process of industrial work practices at SMKN 1 Cikande. By using a website and the PHP programming language, this information system can be used anytime and anywhere. The Waterfall method is a method that implements design by working with users. By using the Waterfall Method, the system can be designed according to the needs and desires of the user. The design results show that the system can record the registration process and ongoing industrial work practice guidance, both to students and supervisors who use it. This design is very useful for the administration and vice principal in providing information and integrating the results of industrial work practices at SMKN 1 Cikande.
This study aims to describe the various kinds of language styles in surah al-Zalzalah. This research is classified as a qualitative research and it uses descriptive methods by describing as well as analyzing the data in surah al-Zalzalah. Meanwhile, it applied stylistic approach which aims to explore the characteristics of the text based on the language style used in surah al-Zalzalah. In addition, the stylistic approach is used to obtain hidden meanings in surah al-Zalzalah so that the minor and major themes of the surah are revealed. This research confirms the existence of rhetorical language in various forms of language style spread in surah al-Zalzalah which are able to have greater influence and effect on the hearts and minds of the readers. From this research, it is concluded that there are twelve language styles in surah al-Zalzalah that are grouped into three stylistic levels. Among those twelve styles, three language styles are at the phonological level (al-mustawÄ al-sawti), four language styles are at the syntactic level, and five language styles are at the imagery level (al-mustawÄ al-taá¹£wÄ«ri).
