Blastocystis sp. is a protozoan parasite, commonly found in the gastrointestinal tracts of animals and humans globally. The parasitic species has wide genetic diversity. Currently the mammalian and avian isolates of the parasite are grouped into 17 well known subtypes (STs), of which ten (ST1-ST9, ST12) are reported in humans. To assess the genetic diversity of Blastocystis sp. in wildlife, a total of 200 fresh fecal samples were collected from 32 mammalian wildlife species in Bangladesh National Zoo. Blastocystis sp. was screened and subtyped by PCR amplification and sequencing of the small subunit ribosomal RNA (SSU rRNA) gene. The minimum prevalence of Blastocystis sp. infection was 15.5% (31/200) in zoo animals. Eight out of 32 wildlife animal species (25.0%) were infected with Blastocystis sp. Among them, the occurrence of Blastocystis sp. was higher in non-human primates (NHPs) (31.8%) than that in herbivores (4.9%) and carnivores (0). Nucleotide sequence analysis of the SSU rRNA gene revealed seven different Blastocystis sp. subtypes, such as ST1, ST2, ST3, ST10, ST11, ST13 and ST14 in the wild animals. ST3 was the dominant subtype (41.9%, 13/31) being detected in NHPs. Of the 31 Blastocystis sp. isolates from the wild animals, 24 (77.4%) isolates belonged to the most common subtypes (ST1 to ST3) found in humans. This is the first molecular study of Blastocystis sp. in wild animals in Bangladesh. This study highlights the remarkable genetic diversity in Blastocystis sp. isolates from zoo animals and provides the first molecular evidence from spotted deer, gayal and grey langur. Due to circulation of large percentage of potentially zoonotic subtypes in the wild animals, there is a higher risk of zoonotic transmission of Blastocystis sp. in the zoo keepers and visitors.
Subclinical mastitis is an economically important disease of dairy cows and has a prominent place amongst the factors that limit milk production. This study was undertaken to determine the association of somatic cell counts (SCC) and occurrence of bacteria with SCM in smallholder dairy cows in Mymensingh, Bangladesh. A total number of 240 quarters milk samples from apparently healthy lactating cows were subjected to SCC using NucleoCounter ® SCC-100 ™ (Chemo Metec). A quarter was considered SCM positive if the quarter had SCC>100 x 10 3 cells/ml. All subclinical mastitis positive quarter milk samples were subjected to bacteriological examination and isolates were classified into major, minor, uncommon and mixed pathogens. The overall quarter-level prevalence of subclinical mastitis of dairy cows in Mymensingh district was 25% (95% CI, 19.52% to 30.48%). The most frequently isolated bacterial species were Staphylococcus aureus (18.33%) followed by coagulasenegative staphylococci (10%), Enterobacter spp. (6.67%), Escherichia coli (5%), Bacillus spp. (5%) and Pseudomonas aeruginosa (5%). Different bacterial isolates were associated with 90% cases of subclinical mastitis as mono infections or mixed infections. Mono and mixed infections significantly influenced SCC and were the most prominent factors responsible for increasing SCC. Background and Aim: Subclinical mastitis (SCM) is one of the most prevalent diseases of dairy cows, and somatic cell count (SCC) is widely used to determine SCM and milk quality. However, setting the threshold of SCC is very important. This study aimed to determine the cow-level threshold of SCC to differentiate milk of SCM affected cows from normal milk in Bangladesh. Materials and Methods: Sensitivity (Se) and specificity (Sp) along with other characteristics of different thresholds of SCC were determined considering the bacterial culture as the gold standard test. Three definitions of intramammary infection (IMI) were set based on the group of pathogens involved. Five categories of SCC thresholds were considered for analysis. Results: Se and Sp of thresholds of SCC greatly varied as definitions of IMI changed. Irrespective of SCC thresholds, Se increased when isolation of major pathogens included in IMI definitions. Se decreased when SCC thresholds increased (from 100 to 300 × 103 cells/mL) for all IMI definitions and ranged from 60.6% to 20.3%. The highest Se was found at low SCC threshold (100 × 103 cells/mL), which resulted in less false-negative outcome. On the other hand, Sp increased with the increment of SCC thresholds giving rise to a less false-positive outcome. Conclusion: The cow-level SCC threshold of 100 × 103 cells/mL was found appropriate to identify subclinical IMI of dairy cows.
Avian influenza (AI) could have potential public health threat in Bangladesh. The present study aimed to acquire the information on knowledge, attitude and practices (KAP) of the backyard and commercial poultry farmers, farm workers, live bird seller, live bird market workers and transporters, and identify risk factors related to KAP on poultry rearing and other practices pertaining to AI. This survey was conducted in Gazipur, Mymensingh (AI reported districts) and Habiganj (AI nonaffected district). A total number of 600 respondents were interviewed with a structured questionnaire prepared in Magpi and data were collected using Android smart phone. More than half (55.7%) respondents had heard about AI and mass media was the predominant source of information (46.7%). A total of 60.2% of the respondents agreed that AI is a serious and 65.7% agreed that it is a preventable disease. Regarding hygienic practices 32% respondents use disinfectant regularly and 73.2% respondents wash hands after handling of birds. Protective measures such as the use of gloves and mask were not used by 93% respondents. The results indicated a higher knowledge score among the respondents of affected areas than those of nonaffected areas. Regarding the attitude score, it was revealed that 54.5% of respondents had good attitude scores towards AI. That is understood that the majority of respondents had positive attitude towards AI. Regarding the practices score, it revealed that 49.5% had practice at a satisfactory level. The practice score was more satisfactory among the respondents of affected areas than those of non-affected areas. The information on levels of knowledge, attitude and practices regarding AI gathered in this study could provide scientific support to assist the Bangladesh government in developing strategies and health education campaigns to prevent transmission of the AI virus among the general population.
