The thermal stability increase of a-amylase obtained from locale bacteria isolate Bacillus subtilis ITBCCB148 was achieved by immobilization process using an ionic exchange matrix of DEAE-Cellulose. The result showed that the immobilized enzyme has an optimum temperature of 60°C; KM 14.8 mL substrate and Vmax 42.4 U/mL. The thermal stability storage temperature of 60°C, pH 9.0 and 60 minutes demonstrated the immobilized enzyme has residual activity of 28.1%; ki = 0.0224 min.-1; and ΔGi = 103.7 kJ mol-1. Although the immobilized enzyme’s thermal stability was only increased 1.5 times, at higher temperatures, it was much more stable than the native enzyme.
Artocarpus kemando Miq. is a species from the family Moraceae in Indonesia known as the Pudau plant. The purpose of this study is to isolate, characterize, modify and test the antibacterial activity of artocarpin compounds isolated from pudding plants. Stages of research include sample preparation, extraction, isolation, and purification of compounds using chromatographic techniques (TLC, VLC, and CC), characterization of compounds using spectroscopy. The isolated compound was obtained as a yellow crystal with a melting point of 185-187 °C. Based on the results of spectroscopic analysis, showed that the prenylated flavonoids had been successfully isolated, artocarpin in an amount of 35.2 mg. Modification of artocarpin with acetic anhydride produces artocarpin acetate as a yellow crystal. Artocarpin and modified compounds showed antibacterial activity against Bacillus subtilis and Escherichia coli.
The derivative of flavonoid compounds, artocarpin (1) and artonin M (2), were isolated from the root wood of Artocarpus altilis and from the root bark of A. kemando, respectively. Both plants originated from Lampung, Indonesia. The structure of the two compounds has been carefully determined by physical method and spectroscopy techniques of UV, IR, and NMR. The in vivo antimalarial test of artocarpin showed very good Plasmodium activity in female mice, with ED50 value of 34.88 mg/kg body weight (kgBW), whereas the in vitro antimalarial test of artonin M showed very strong activity with IC50 of 0.3 μg/mL (5.967 x 10-7 M).
Production, purification and characterization of the α-amylase from local bacteria isolate Bacillus subtilis ITBCCB148 has been successfully carried out. The purified enzyme increased 149.9 times with specific activity of 115,500 U/mg compared to enzyme crude extracts. This enzyme has an optimum pH of 5.5 and an optimum temperature of 60°C and can maintain its stability at a temperature of 60-80°C, thus this enzyme is categorized as a thermostable enzyme.
Salah satu tugas Tri Dharma Perguruan Tinggi adalah melakukan pengabdian kepada masyarakat, di samping pengajaran dan penelitian tentu saja. Kegiatan pengabdian ini dilaksanakan di Dinas Ketenagakerjaan dan Transmigrasi Kabupaten Sijunjung Sumatera Barat. Tema yang diangkat adalah Pelatihan Batik dan Manajemen Kewirausahaan. Metode pelaksanaan kegiatan dilakukan dengan ceramah, demonstrasi, dan pelatihan. Peserta pelatihan difokuskan pada masyarakat yang sedang memulai usaha kecil dan menengah, serta warga masyarakat yang terdampak baik secara langsung dan tidak langsung wabah COVID-19. Pada akhirnya pelatihan ini memang tidak bisa hanya berhenti di sini saja karena aplikasi secara langsung bagaimana membatik sangatlah diperlukan baik itu tekhik batik tulis maupun cap.
