A new sensitive determination method of estradiol in plasma using peroxyoxalate ester chemiluminescence combined with an HPLC system

Abstract: A new sensitive determination method of estradiol in a plasma sample using peroxyoxalate ester chemiluminescence was developed. Estradiol, which was extracted by liquid-liquid extraction using ethyl acetate from plasma, was derivatized with dansyl-chloride (DNS-Cl) and separated by reverse-phase HPLC. The performance of four oxalates, bis(trichlorophenyl)oxalate (TCPO), bis(2,4-dinitrophenyl)oxalate (DNPO), bis(pentafluorophenyl)oxalate (PFPO), and bis[4-nitro-2-(3,6, 9-trioxadecyloxycarbonyl)phenyl] oxalate (… Show more

“…Recovery experiments were carried out by spiking plasma samples with different concentrations of standard vitamin K homologues, founding values ranging from 82.0% to 93.5% for all vitamin K homologues. The PO system has also shown its usefulness in the detection of a steroid hormone, estradiol, in plasma samples [58]. Estradiol was extracted by liquid-liquid extraction, and derivatized with dansyl-chloride.…”

“…Due to its high selectivity and sensitivity and its wide linear working range, HPLC-CL analysis has become an attractive technique for pharmaceutical and biomedical analyses, [12][13][14][15] but not for determination of environmental samples. Generally, CL analysis can be conducted based on the CL induced by the reactions of an analyte with CL reagents, or the inhibition/sensitization of CL which result from the reaction of the analyte with the CL reagents or oxidants prior to the CL reaction.…”

“…The earliest analytical form of the liquid‐phase CL method [17] has received considerable attention due to its advantages of excellent sensitivity, low reagent consumption, and effective avoidance of reagent dilution associated with flow, static injection methods. However, most of the previously static injection methods have adopted the manual injection mode, which may generate poor repeatability results under the injection speed, which is highly dependent on the operator [18]. To further improve the poor reliability of manual injection, a modification in conjunction with other precise injection devices was envisaged.…”
