In this study, piperine was investigated as a drug molecule due to its analgesic and anti‐inflammatory properties. Piperine was encapsulated with poly(lactic‐co‐glycolic acid) (PLGA) in order to increase drug bioavailability for topical applications. Characterization of piperine PLGA nanoparticles (NPs) were performed with spectroscopic and imaging methods. It was found that the piperine PLGA NPs were successfully prepared according to the analyses. The encapsulation efficiency and loading capacity of the piperine were calculated as 99% and 3%, respectively. In vitro release study was also performed. Besides, it was shown that piperine had no mutagenic effect. Finally, molecular docking simulation was performed by using the AutoDock Vina program to identify the interaction between piperine and COX‐1 and COX‐2 enzymes. The aim of this study is to contribute to the design of plant derived nano‐formulations for local anti‐inflammation and pain treatment with better skin penetration.

Congenital cataracts are associated with gene mutations, yet the underlying mechanism remains largely unknown. Here we reported an embryonic chick lens model that closely recapitulates the process of cataract formation. We adopted dominant-negative site mutations that cause congenital cataracts, connexin, Cx50E48K, aquaporin 0, AQP0R33C, αA-crystallin, CRYAA R12C and R54C. The recombinant retroviruses containing these mutants were microinjected into the occlusive lumen of chick lenses at early embryonic development. Cx50E48K expression developed cataracts associated with disorganized nuclei and enlarged extracellular spaces. Expression of AQP0R33C resulted in cortical cataracts, enlarged extracellular spaces and distorted fiber cell organization. αA crystallin mutations distorted lens light transmission and increased crystalline protein aggregation. Together, retroviral expression of congenital mutant genes in embryonic chick lenses closely mimics characteristics of human congenital cataracts. This model will provide an effective, reliable in vivo system to investigate the development and underlying mechanism of cataracts and other genetic diseases.

Piperine and capsaicin are important molecules with biological and pharmacological activities. This study aimed to evaluate the cytogenotoxic and protective effect of piperine and capsaicin on Allium cepa cells. A. cepa roots were exposed to negative (2% Dimethylsulfoxide) and positive (Methylmethanesulfonate, MMS, 10 µg/ mL) controls, and four concentrations (25-200 µM) of piperine or capsaicin (alone) or associated before, simultaneously or after with the MMS. Only the lowest concentration of piperine (25 µM) showed a protective effect because it was not genotoxic. Piperine and capsaicin were cytotoxic (50, 100 and 200 µM). Piperine (50 to 200 µM) caused a signifi cant increase in the total average of chromosomal alterations of in A. cepa cells.For capsaicin, the genotoxic effect was dose-dependent with a signifi cant increase for all concentrations, highlighting the signifi cant presence of micronuclei and nuclear buds for the two isolates. In general, bioactive compounds reduced the total average of chromosomal alterations against damage caused by MMS, mainly micronuclei and/or nuclear buds. Therefore, the two molecules were cytotoxic and genotoxic at the highest concentrations, and did not have cytoprotective action, and the lowest concentration of piperine demonstrated important chemopreventive activity.
