Cell-bound IgE and plasma IgE as a combined clinical diagnostic indicator for allergic patients

Abstract: Allergic responses are mainly caused by IgE, which is often located on the cell surface. The current diagnostic method detects both allergen-specific IgE and total IgE levels, but a number of allergic patients have a normal serum IgE level, which is a poor clinical correlate for allergy. Here, we developed a simple method to detect the level of cell-bound IgE by dissociating it from blood cells with lactic acid. Dissociated cell-bound IgE and plasma IgE levels were detected using the same ELISA kit at the same… Show more

“…The positive HDM component IgE number was also higher in the AR group. Although there may be discordance in serum IgE level and cell-bound IgE level in some individuals, in general, serum IgE correlates well with cell-bound IgE on FcεRI+ cells [33][34][35]. Thus, higher level of serum HDM and Der p1/p2 IgE implies higher density of cell-bound IgE on the surface of mast cells and basophils, which facilitates the binding of HDM to IgE, crosslinks the IgE antibodies on the cell surface, and then triggers downstream mediator release.…”

“…Ours results also suggests that the ex vivo challenge LAIT Despite the free serum IgE correlates well with allergic disease, there are some allergic patients that present normal IgE serum levels at immunoassays. That is why it is actually proposed the measuring, by the employ of lactic acid on plasma, of the liberated cell bounded IgE as a tool to help physicians to diagnosis allergy [61]. This may happen when the allergy is produced by a non-IgEmediated mechanism, or even when the serum presence of non-IgE antibodies interferes with the IgE immunoassay, competing by the allergens' epitopes [62].…”

“…In atopic individuals, total IgE levels tend to be increased [ 25 ]. This feature has also been found in some cohorts of SLE patients; several groups studied the relationship between atopy and SLE with some controversial results [ 26 , 27 , 28 , 29 , 30 ].…”
