Comparison of antidiabetic potential of (+) and (−)-hopeaphenol, a pair of enantiomers isolated from Ampelocissus indica (L.) and Vateria indica Linn., with respect to inhibition of digestive enzymes and induction of glucose uptake in L6 myotubes

Abstract: The remarkable α-glucosidase inhibition exhibited by the acetone extract of the rhizome of Ampelocissus indica (L.) and stem bark of Vateria indica Linn. (IC50 23.2 and 1.47 μg mL−1) encouraged us to isolate the phytochemicals from these plants.

“…(+) Hopeaphenol and (−) hopeaphenol oligomer (tetramer) of resveratrol isolated from Ampelocissus indica (L.) and Vateria indica Linn., respectively, displayed IC 50 values of 21.21 ± 0.987 and 9.47 ± 0.967 mM in an α-glucosidase inhibitory assay [21], which were higher than the standard acarbose (IC 50 81.3 ± 1.10). The compounds showed a concentrationdependent inhibition of both α-glucosidase and α-amylase enzymes.…”

“…Further, the study also indicated the positive effect of hopeaphenols as antiglycating agents, with IC 50 values of 81.9 ± 1.176 and 50.96 ± 0.897, respectively, for (+) and (−) isomers which again were less than the ascorbic acid standard (IC 50 158.23 ± 0.718). The results indicated that the hopeaphenols can be a promising natural compound in diabetic management [21]. The effect of glucose uptake performed by 2-NBDG in L6 rat skeletal muscle cells using flow cytometry (BD FACS Aria II, USA) showed potent glucose uptake by (+) and (−) hopeaphenol of 31 and 26.4%, respectively [21].…”

“…Flavonoids are reported as antioxidants and scavengers of free radicals (19). The antioxidant activity of A. indica is proven (10).So the avonoids present in the drug A.indica may be the main phytochemical responsible for the observed anti-in ammatory property of the drug.…”

“…A.indica(AI) is locally known as Chembravalli documented in Hortus Malabaricus (5), and some Keraliya traditional books like Chikitsamanjari(6), Arogyakalpadrumam 7, Yogamrutam (8) and Vaidyatarakam(9) prescribed for in ammatory skin ailments, wound healing. Anti oxidant, anti diabetic and diuretic properties of the plant and its fractions has been explored earlier (10,11). Since the traditional knowledge claims the cure of in ammatory skin ailments and wounds; the present study intended to see the anti-in ammatory effect of A.indica in Wistar rats Carrageenan-induced paw edema is one of the most popular and highly sensitive and reproducible test for new non-steroidal anti-in ammatory drugs (12).…”
