Construction of new GFP-tagged fusants for Trichoderma harzianum with enhanced biocontrol activity

Abstract: Abstract:Trichoderma is one of the most exploited biocontrol agents for the management of plant diseases. In biocontrol ecology, the critical factors are detection, and the monitoring and recovery of specific biocontrol agents either naturally present or deliberately released into the environment. Protoplast fusion is an appropriate tool for the improvement of biocontrol Trichoderma strains. Protoplast isolation from Trichoderma harzianum was achieved using 24 h culture age, 6.6 mg/ml Novazym L 1412 at 30°C wh… Show more

“…Trichoderma species are known to produce large quantities of fungi-toxic metabolites. They are the active mycoparasites which have been used as effective biocontrol agents against foliar and soilborne disease, as well as plant parasitic soilborne nematodes [83,158]. Meyer and his colleagues have investigated the use of Trichoderma species against root-knot nematodes on a wide range of crops such as tomato, okra, mungbean, and bell pepper [159].…”

“…T. harzianum was in general the best in reducing incidence compared with B. subtilis. In this respect Kowsari et al, (2014) reported that, analysis of gene expression during these parasitizing events suggests that fungal cell walldegrading enzymes are actively produced by T. harzianum. * 1L:100L water= 1 liter from original solution (30x10 6 CFU/mL) diluted in 100 liter water to give 30x10 4 CFU/mL concentration; 1L:75 L= water= 1 liter from original solution (30x10 6 ) diluted in 75 liter water to give 40x10 4 CFU/mL concentration; 1L:50 L water= 1 liter from original solution (30x10 6 ) diluted in 50 liter water to give 60x10 4 CFU/mL concentration.…”

“…The protoplast fusion was performed by using the method of Kowsari, Motallebi, and Zamani (2014) with some modifications. 10 mM CaCl 2 , pH 6.5) was used as a fusogen agent.…”
