Development of TaqMan real-time PCR assays for monitoring Vibrio harveyi infection and a plasmid harbored by virulent strains in European abalone Haliotis tuberculata aquaculture

Abstract: International audienceThe Gram-negative bacterium Vibrio harveyi is known to be highly pathogenic for the European abalone Haliotis tuberculata, which is a gastronomically important marine gastropod with a high commercial value. Since 1998, some particular bacterial strains are described as implicated in recurrent mortality outbreaks in French farm and field stocks of abalone. Recently, a 9.6 kb plasmid named pVCR1, was shown to be harbored by one highly V. harveyi virulent ORM4 strain suggesting its involveme… Show more

“…Their purity was verified by real‐time PCR (Schikorski et al . ). BB120 strain was shown to exhibit strong virulence in brine shrimp Artemia franciscana, whereas the use of AI‐2 and CAI‐1‐deficient mutants abolished virulence of V. harveyi to brine shrimp (Defoirdt and Sorgeloos ).…”

“…Liquid broth media screening: The strains used for this bioassay are V. harveyi BB120 wild type and its derived double mutants (JAF 375, JMH 597 and JMH 612) obtained from Bassler's laboratory (Bassler et al 1997;Henke and Bassler 2004a,b). Their purity was verified by real-time PCR (Schikorski et al 2013). BB120 strain was shown to exhibit strong virulence in brine shrimp Artemia franciscana, whereas the use of AI-2 and CAI-1-deficient mutants abolished virulence of V. harveyi to brine shrimp (Defoirdt and Sorgeloos 2012).…”

“…In addition, for V. harveyi-related species, highly similar genomes and genome plasticity may also limit precise identification by molecular techniques in some cases (Thompson and Swings, 2006;Sawabe et al, 2007). Thompson et al, 2007;Cano-Gomez et al, 2011), loop-mediated isothermal amplification (LAMP) (Cao et al, 2010), and TaqMan PCR to detect potentially virulent strains for H. tuberculata (Schikorski et al, 2014) (Table 2).…”

“…In the current study, we have used and tested a TaqMan probe developed by Schikorski et al (2013) for qPCR assays of V. harveyi (16). Measurements obtained by TaqMan qPCRs following a scheme of serial dilution of a pure culture of V. harveyi in samples of hemolymph or tissues showed an excellent linear correlation, and TaqMan qPCRs have been proven to be powerful and sensitive techniques for V. harveyi detection and quantification in European abalones.…”
