Direct and indirect detection of 2n eggs in hybrid diploid families derived from haploid tbr × wild species crosses

Abstract: The analytic-synthetic scheme makes use of haploidization and 2n gametes-mediated sexual polyploidization to incorporate genetic diversity from wild related species in potato breeding. Whereas 2n pollen can be directly screened by microscopy, 2n eggs are routinely screened indirectly by interploid crosses, a timeconsuming and inaccurate technique. In five haploid tbr · wild diploid species families with variability for agronomic characters, 2n eggs were screened indirectly to incorporate this germplasm into a … Show more

“…The diameter of spherical 2n pollen is, in theory, 1.26 times that of normal reduced pollen (Erazzú and Camadro 2007). The 2n pollen of Ipomoea trifida (Orjeda et al 1990).…”

“…M6 was screened for the presence of 2 n eggs by crossing it as a female to tetraploid cultivars. Due to a triploid block in potato, seeds are produced from 2 x by 4 x crosses only if 2 n eggs are present (Erazzú and Camadro, 2006).…”

“…However, it produces approximately one seed per pollination when crossed as a female to tetraploid clones. The production of hybrid seeds from crosses to tetraploids indicates that M6 produces some 2 n eggs (Erazzú & Camadro, 2006).…”

“…The frequency of 2n egg formation has often been estimated after crosses between 2x x 4x plants. A cross between a diploid seed parent and tetraploid pollen parent will only result in good tetraploid seed when 2n egg cells are present (Conicella et al, 1991;De Haan et al, 1992;Erazzu & Camadro, 2007;Estrada-Luna et al, 2004;Jongedijk, 1987;Lamote et al, 2002;Ortiz & Peloquin, 1991;Veronesi et al, 1986;Van Laere et al, 2009;Werner & Peloquin, 1987). In Triticum-Aegilops hybrids, Zhang et al (2007a used the selfed seedset as a good indication for the formation of 2n gametes.…”
