Direct fluorogenic detection of palladium and platinum organometallic complexes with proteins and nucleic acids in polyacrylamide gels

Abstract: Allyl-and propargyl ethers of umbelliferone are sensitive probes for palladium and platinum, including anticancer compounds cisplatin, carboplatin and oxaliplatin, and effective for direct visualization of protein and DNA complexes with organometallic compounds in polyacrylamide gels allowing easy detection of interactions with analyzed protein or nucleic acid. Both probes can be used for fast evaluation of Pd/Pt binding to nanocarriers relevant in drug targeted therapy or specific clinically relevant target m… Show more

“…This was because SBF-Pg and SBF-2 × Pg were almost insoluble in pure water, and in previous experiments, we have noticed that the response to Pd and other metals is strongly influenced by a solvent. 15 All synthesized SBF probes are virtually non-fluorescent, but upon contact with Pd in the presence of sodium borohydride (BH), the probes are rapidly deprotected into corresponding phenolates (Fig. 1b), yielding blue or yellow fluorescence (Fig.…”

“…11 Using these strategies, several probes have been designed to effectively detect Pd complexes in solutions, 12,13 in cells 14 or in complexes with biomolecules. 15 In this work, we have designed and synthesized a color tunable SBF sensor for sensitive detection of Pd in solution. In addition, in this study, we demonstrate that our synthesised SBF sensor can be used for a facile, high-throughput analysis of bioorthogonal reactivity of Pd-based catalysts designed for bioorganometallic reactions in physiological milieu.…”

“…For this purpose, we employed our recently developed technique for the detection of protein-organometallic compound interactions in native PAGE, utilizing the palladium-catalyzed Tsuji-Trost reaction to convert nonfluorescent umbelliferone propargyl ether to UV-excitable umbelliferone. 37 The technique is based on separation of the protein/organometallic complexes in native PAGE. The gel is then soaked in an aqueous solution of propargyloxy umbelliferone, washed, treated with a solution of sodium borohydride, and observed under a transilluminator.…”
