Efficacy of Different Collection Techniques on Yield and Quality of Egyptian Buffalo Oocytes.

Abstract: Buffaloes ovaries obtained from slaughterhouses were used to study the influence of the oocyte collection techniques (dissection, aspiration, slicing, and aspiration plus slicing) on the availability of oocytes quantity and quality of buffalo oocytes. The oocytes were collected aseptically from the ovaries by the four methods. In all methods, oocytes were classified into 5 classes on the basis of the morphology of compact, denuded, degenerated, expanded and partial denuded oocytes. Results showed that the oocy… Show more

“…Results showed that; bovine oocytes maturation time is about 28-30hrs., which is correlated to the collection method in which the aspiration method yield a less oocytes count but with a predominant degree of maturation in regarding to other methods. (34) observed that; when trying to compare between different methods for oocytes collection in regarding to the Oocytes number, quality and degree of maturation, the slicing and slicing after aspiration methods came in earlier methods that yield more Oocytes count with good quality that reached to maturity condition in well considered degree. In present study we conclude that; kind of bovine oocytes collection from abattoir ovarian specimens may have an effect on oocytes count as well as maturation.…”

“…Ovaries which do not possess CL have higher number of follicles being recovered and also higher grade COCs than those non CL bearing ovaries (Khandoker et al, 2011). Furthermore, the stroma of buffalo ovaries are thick and follicles are deeply embedded in them and therefore aspiration of oocytes from follicles gets difficult (Selvaraj et al, 2008, Hammad et al, 2014. The ovaries which had CL yielded lower number of oocytes as compared to non CL bearing ovary as it was discussed by (Kumar et al 1997).…”

“…In vitro maturation is a method in which immature oocytes of dead animals are retrieved from the ovary using various methods such as aspiration, scoring, slicing, puncture or a combination of aspiration plus slicing method (Khan et al, 1997, Mehmood et al, 2011, Hammad et al, 2014 and are allowed to mature in the laboratory which is a crutial step for fertilization and subsequent embryonic development. Maturation rate in cattle and buffaloes is almost similar (94 v/s 87% respectively) but a slightly lower cleavage rate is observed in buffalo as compared to cattle (84 v/s 65%) (Gasparrini, 2002, Neglia, 2003 In vitro fertilization has drawn interest of many researchers as it can salvage the genetic potential from infertile female and can also yield large number of embryos from slaughter house derived oocytes which may go waste otherwise (Kumar 2012).…”

“…According to Hammad et al (2016) when trying to compare between different methods for oocytes collection in regarding to the oocytes number, quality and degree of maturation, the slicing and slicing after aspiration methods came in earlier methods that yield more oocytes count with good quality that reached to maturity condition in well considered degree. The oocytes collection methods also did not influence subsequent embryonic developmental competence after in vitro fertilization with M II stage oocytes (Wang et al, 2007).…”
