Formation and properties of 1000-A-diameter, single-bilayer phospholipid vesicles.

Abstract: Two methods are reported for the formation of large, uniform-sized phospholipid vesicles. (5) or ether injection (6). Vesicles may be formed with phospholipid mixtures, purified phospholipids, or synthetic phospholipids. The methods of preparation and the properties of such vesicles have recently been reviewed (1, 7). Although multilamellar liposomes are easy to prepare, they have been criticized as a model system for the study of membrane phenomena (1). Thus, the kinetics of transport processes are more comp… Show more

“…LUVs of 100 nm diameter were prepared from a thoroughly dried phospholipid film of typically 18 mg, according to the method described by Enoch and Strittmatter with slight modifications [22]. The lipid film was suspended in a NaC1/Tris-aeetat~ buffer (100 mM NaCI/20 mM Tris-acetate (pH 8.1)) containing 10 mM deoxycholate in a lipid/deoxycholate molar ratio of 2.…”

“…(approx. 0 °C for 10 mol~ DPPC [30]), a prerequisite in the vesicle preparation technique used [22]. Moreover, it is to be expected that vesicle size and fluidity are not much affected on substituting DPPC by DHPC at this concentration [27,31].…”

“…As determined by measurement of the number of lipid molecules per vesicle, there are approximately 3000 strands of PEG 2000 molecules attached to the liposome surface and about 1% of these PEG strands are conjugated with HuCC49 Fab 0 fragments. 13,25 Nuclease treatment was able to remove any pDNA, either free or bound to the exterior of liposomes. Ethidium bromide staining after agarose gel electrophoresis demonstrated complete removal of any surface-bound pDNA from the preparation.…”
