GC–MS metabolomics revealed protocatechuic acid as a cytotoxic and apoptosis-inducing compound from black rice brans

Abstract: GC-MS metabolomics was used to discriminate the phytochemicals profile of Indonesian white, red, and black rice brans, and Japanese white rice brans. This technique was used for the first time to identify compounds in rice brans having cytotoxic activity against WiDr colon cancer cells. Orthogonal Projection to the Latent Structure (OPLS) analysis showed that protocatechuic acid (PA) was a discriminating factor found in black rice brans which strongly correlated with its cytotoxicity (IC. ±. µM). Real time-PCR… Show more

“…MDA-MB-231 is a more aggressive late-stage model of breast cancer, which would explain the difference in activity observed between MCF-7 and MDA-MB-231. Contrary to our results, a previous study with protocatechuic acid from black rice bran has indicated that purified protocatechuic acid can induce apoptosis in cells even with range of low concentrations such as 10-50 µg/ml (20). Similarly, numerous studies have reported the effect of procyanidin B type dimer and trimers to be effective against cancer cells (22,23,41).…”

“…A human study demonstrated an increase in antioxidant activity even with a single dose of date seed products such as DSE, date seed powder (DSP), and date seed supplemented Arabic pita bread (DSB) (18). Moreover, many polyphenols such as catechin, epicatechin, p-hydroxybenzoic acid, protocatechuic acid, and procyanidins have reported anti-tumour property (19)(20)(21)(22)(23)(24). Although these compounds are abundant in DS, there is no detailed investigation of the anti-tumour potential of DS.…”

“…Histopathological examination commonly contains immature tissue in various amounts. Most common are neuroectodermal (16) tubules and rosettes, but occasionally with active glial cell components, mitosis in significant amounts mixed with ectodermal and endodermal elements with varying levels of maturation. Neuroectodermal tubules are coated by hyperchromatic cells arranged in a stacked way with many mitose and may also contain pigments.…”

“…At the same dose level, it also activated the extrinsic pathway by modulating caspase-8. On the other hand, PCA showed positive regulation of caspase-1 and caspase-7 at higher doses (25-50 µg/mL), suggesting its possible involvement in proinflammatory cell death [79].…”
