Hongjingtian Injection Attenuates Myocardial Oxidative Damage via Promoting Autophagy and Inhibiting Apoptosis

Abstract: Natural products with antioxidative activities are widely applied to prevent and treat various oxidative stress related diseases, including ischemic heart disease. However, the cellular and molecular mechanisms of those therapies are still needed to be illustrated. In this study, we characterized the cardioprotective effects of Hongjingtian Injection (HJT), an extensively used botanical drug for treating coronary heart disease. The H/R-induced profound elevation of oxidative stress was suppressed by HJT. HJT a… Show more

“…Surgical Procedure MIRI was surgically performed according to previously described methods. 18) Rats were anesthetized intraperitoneally using 5% chloral hydrate (6 mL/kg, Beijing Solarbio Science & Technology Co., Ltd., China). MIRI was induced by left thoracic incision to expose the heart and a 5/0 silk suture was sewn around the left anterior descending coronary artery.…”

“…Specifically, it has been shown to dilate the coronary artery and reduce cardiac afterload in canine models (Zhang et al, ). It has also demonstrated cardioprotective effects against oxidative stress both in hypoxia/reoxygenation (H/R) ‐induced myocardial injured cells and in myocardial ischemia‐injured rats (Zhang et al, ).…”

“…All cells were maintained in 5% CO 2 at 37°C under a humidified atmosphere. After serum starvation for 24 hrs, cells were exposed to DMEM containing 5.5 mmol/L D-glucose +19.5 mmol/L D-mannitol (MG), 25 mmol/L D-glucose (HG), 25 mmol/L D- glucose + 10 mL/L HJT(HG + HJT10), 25 mmol/L D- glucose + 20 mL/L HJT(HG + HJT20), or 25 mmol/L D- glucose + 40 mL/L HJT (HG + HJT40) 15. For the inhibition experiments, cells were divided into five groups: MG, HG, HG + LY294002, HG + HJT40, HG + HJT40 + LY294002.…”
