Human salivary protein extraction from RNAPro·SAL™, Pure·SAL™, and passive drooling method

Abstract: Objective:The aim of the current study was to carry out a preliminary validation of devices for standardized collection of whole mouth fluid (WMF) in comparison to the passive drooling method for protein analysis in healthy subjects.Materials and Methods:A carefully designed sample collection/pretreatment protocol is crucial to the success of any saliva proteomics project. In this study, WMF was collected from healthy volunteers (n = 10, ages: 18–26 years). Individuals with any oral disease were excluded from … Show more

“…Tear, plasma, and saliva samples from both healthy and KC participants were processed for western blot analysis, as previously described (Chertov et al, 2004;Chevalier et al, 2007;Khurshid et al, 2017;Priyadarsini et al, 2014). Briefly, Bradford assay (Thermo scientific, IL, USA) was carried out to determine the protein concentration and purity of saliva and plasma samples, though not to tear samples due to small volume (~5ul).…”

“… 24 25 26 Saliva contains a significant number of biomarkers and components such as RNA and DNA, various immunoglobulins, microorganisms, and metabolites. 27 Saliva sample collection is also considered to be cheap, easy, and of minimal risk of transmitting the disease to health care personnel as self-collection involving only the patient is possible. 23 Human salivary glands are reservoir for the viruses and most likely cell receptor for SARS-CoV-2 investigated in the serological examinations of patient samples concurred to be the angiotensin-converting enzyme 2 (ACE-2) receptor expression.…”
