Identification of anti-HIV and anti-Reverse Transcriptase activity from Tetracera scandens

Abstract: We report here that an ethanol extract of Tetracera scandens, a Vietnamese medicinal plant, has anti-HIV activity and possesses strong inhibitory activity against HIV-1 reverse transcriptase (RTase). Using a MT-4 cell-based assay, we found that the T. scandens extract inhibited effectively HIV virus replication with an IC50 value in the range of 2.0-2.5 μg/ml while the cellular toxicity value (CC50) was more than 40-50 μg/ml concentration, thus yielding a minimum specificity index of 20-fold. Moreover, the ant… Show more

“…Following these two steps, the host cell produces viral proteins and genomic material using proteases. Studies with natural products with anti-HIV activity focus on anti-retroviral drugs that inhibit the activity of these three types of enzymes: RT, proteases and integrases (Kwon et al, 2012;Suedee et al, 2014).…”

“…This activity is related to the presence of calanolides, but the plant source is relatively rare. Kwon et al (2012), studying Tetracera scandens (Dilleniaceae) extracts, estimated an IC 50 level of 0.7 μg mL -1 against in vitro HIV-1 RT. According to García-Zebadúa et al (2011), considering the HIV-1 RT inhibitory properties of plant extracts, some authors have proposed that an IC 50 ≤ 50 μg mL -1 may be considered potent.…”

“…Relative amounts of LDH secreted to the medium was calculated as LDH present in media/(LDH present in media + LDH present in cell pellet) and compared to that of the control group which was treated with DMSO. An MTT assay was performed as described (30).…”

“…The ethanol extract of T. scandens has anti-HIV activity and high inhibitory activity against HIV-1 reverse transcriptase activity [5]. The methanol extract of T. Scandens stem yields new nor-lupane triterpene capable of exhibiting significant concentration-dependent xanthine oxidase inhibitory activity [1].…”
