“…The SDS-PAGE is a promising tool for distinguishing cultivars of a particular crop species (Cooke, 1984;Gardiner and Forde, 1988). The present study was therefore undertaken in order to distinguish among the five given cultivars of mungbean through the use of SDS-PAGE.…”

“…A bulk sample is a composite seed sample that represents the mixture of electrophoretic phenotypes present in a cultivar. For bulk extraction, a set of at least 200 seeds (Gardiner & Forde, 1992) was used to obtain a representative sample, although our results showed that a batch of 50 seeds provided a reproducible banding pattern (results not shown). Electrophoregrams of the bulk samples were evaluated by the presence/absence of bands and by their intensities, distinguishing four levels from 1 to 4, corresponding to increasing band intensity.…”

“…As a result, new procedures are needed to identify cultivars (14). Doyle et al (11) has successfully used DNA data in grass taxonomy and phylogeny.…”

“…Grass seed storage proteins have already been investigated systematically by several authors (16,26). Seed protein electrophoresis was found to be a useful technique for cultivar discrimination in several economically important grass species including Kentucky bluegrass (Poa pratensis l.) and ryegrass (L. perenne l. and L. multiflorum l.) (8,12,13,14,15,20,21,28). it is possible to identify and certify individual genotypes to analyze the progenies and populations (2,18,24,27).…”
