Improvement of gastroprotective and anti-ulcer effect of kenaf seed oil-in-water nanoemulsions in rats

Abstract: Kenaf seed oil-in-water nanoemulsions (KSON) and kenaf seed oil-in-water macroemulsions were produced to access their gastroprotective effect against indomethacin- and ethanol-induced ulcers in comparison with non-emulsified kenaf seed oil (KSO). Emulsifier mixture (EM) that used to emulsify KSO was also included in the study. Ulcer index, stomach tissue oxidative status, and histopathological changes in indomethacin-induced and ethanol-induced ulcer models were both evaluated. KSON had demonstrated good gastr… Show more

“…NSAIDs are the most common prescribed medications for patients suffering from pain and inflammation. They are commonly prescribed for patients over 65 years old with an approximate prevalence of 7.3% [ 6 , 7 , 8 ]. NSAIDs work as anti-inflammatory agents through the inhibition of cyclooxygenase (COX) enzymes.…”

“…Kenaf seed oil‐in‐water nanoemulsions (KSON) stabilized by complexation of beta‐cyclodextrin (β‐CD) with sodium caseinate (SC) and Tween 20 (T20) have been shown to have good lipolysis rate (85.3%) and higher bioaccessibility of vitamin E and total phenolic content than nonemulsified KSO in the in vitro study (Cheong, Tan, & Nyam, ). In a recent study, nanoemulsion form of kenaf seed oil demonstrated significantly better gastroprotective and antiulcer effect on ethanol‐ and indomethacin‐induced acute gastric lesions, compared to nonemulsified kenaf seed oil (Cheong et al., ). It was hypothesized that small nanosized particles of stable kenaf seed oil‐in‐water nanoemulsions could improve the rate of bioavailability as to exert faster beneficial biological effects, compared to nonemulsified kenaf seed oil.…”

“…These results are comparable to those of Cheong et al where the negative control group showed extensive damage of the gastric mucosa, while that treated with omeprazole was subjected to edema and leukocyte infiltration. 34 Interestingly, the gastric tissue of the limonene-based ODTs treated group demonstrated almost normal mucosa with minor inflammation and submucosal hemorrhage ( Figure 4E ). This protection is attributed to the marked gastroprotective activity of limonene against ulcer, which is mediated by inhibition of cytokines, increase the mucus level as well as vasoactive intestinal peptides, decrease ulcer severity, and preserve gastric mucosal integrity.…”

“… 33 The second group, the positive control, received omeprazole (Gastrimut ® ) at an oral dose of 20 mg/kg. 34 The third group was pretreated orally with 10 mg/kg tadalafil aqueous suspension. The fourth group received tadalafil-loaded limonene-based SNES ODT dispersed in normal saline to obtain an equivalent dose, 35 while the fifth group received an equal volume of unloaded limonene-based SNES ODT also dispersed in normal saline.…”
