Lipidation increases antiviral activities of coronavirus fusion-inhibiting peptides

Abstract: Coronaviruses (CoVs) can cause life-threatening respiratory diseases. Their infectious entry requires viral spike (S) proteins, which attach to cell receptors, undergo proteolytic cleavage, and then refold in a process that catalyzes virus-cell membrane fusion. Fusion-inhibiting peptides bind to S proteins, interfere with refolding, and prevent infection. Here we conjugated fusion-inhibiting peptides to various lipids, expecting this to secure peptides onto cell membranes and thereby increase antiviral potenci… Show more

“…Furthermore, since CoV receptors and proteases tend to partition in lipid-enriched domains, another group tested the possibility of directing HR2 peptides towards these lipid-enriched domains where CoV entry occurs. Conjugating palmitate, which partitions into lipid rafts, to HR2P-M2 was found to increase inhibition of SARSpp and MERSpp cell entry (Park and Gallagher, 2017). On the other hand, conjugating tocopherol, which partitions into non-raft domains, to HR2P-M2 had minimal impact on cell entry.…”

“…Besides, the S2 domain of the SARS-CoV S protein contains heptad repeat HR1 and HR2 sequences. Peptide HR2 (HR2: SLTQINTTLLDLTYEMLSLQQVVKALNESYIDLKEL) and its lipid-binding peptide is highly similar or even identical to the near-membrane portion of S protein ferredoxin, which interferes with refolding into post-fusion fusion-catalyzing domains (FDs) ( Du et al, 2009 ; Park and Gallagher, 2017 ). According to recent research, the lipopeptide EK1C4, derived from EK1 (SLDQINVTFLDLEYEMKKLEEAIKKLEESYIDLKEL), is the most effective fusion inhibitor against COVID-19 S protein-mediated membrane fusion ( Xia et al, 2020 ).…”

“…Dectin-1 is a specific receptor for β-glucan, expressed on phagocytes and immunocompetent cells. The investigation of Park and Gallagher (2017) , showed a decrease in the influenza virus replication due to an increase in the interferon-gamma and nitric oxide production. β-glucan is associated with immune-modulatory action like the production of nitric oxide (potent viral replication inhibitor), reactive oxygen species, and release of cytokines ( Reboul, 2017 ).…”
