“…The tomato physical map was used to identify positions for markers linked to yellow leaf curl (Ty) resistance from a wild species introgression (Kadirvel et al 2013), and also for designing markers such as those used in fine mapping a bacterial spot resistance locus derived from a S. pennellii introgression in cultivated tomato (Sharlach et al 2013). Markers were also developed based on the S. pimpinellifolium assembly to map TERMINATING FLOWER (TMF), a gene involved in flowering, and ITAG2.3 coding sequence annotations were used to observe expression of the mapped gene (MacAlister et al 2012).…”

“…Recent studies on tomato leaf curl disease resistant lines derived from S. chilense have shown presence of additional introgressions that confer resistance to ToLCD (Kadirvel et al 2013). It was therefore necessary to test for the presence of these recently reported introgression regions in lines developed in our study.…”

“…In addition, we also used SSR markers reported by Kadirvel et al (2013) to test the possible presence of ty-5 and qTy10.1 in stable Ty-2 and Ty-3 lines developed in this study. For DNA extraction, leaf tissue was collected from young leaves in a 1.5 ml centrifuge tube containing 700 ll of extraction buffer (200 mM Tris-HCl pH 7.5, 250 mM NaCl, 25 mM EDTA, 0.5 % SDS and 5 % freshly added sodium metabisulfite).…”
