Methyl viologen responsive proteome dynamics ofAnabaenasp. strain PCC7120

Abstract: A proteomic approach was employed to elucidate the response of an agriculturally important microbe, Anabaena sp. strain PCC7120, to methyl viologen (MV). Exposure to 2 μM MV caused 50% lethality (LD50 ) within 6 h and modified the cellular levels of several proteins. About 31 proteins increased in abundance and 24 proteins decreased in abundance, while 55 proteins showed only a minor change in abundance. Of these, 103 proteins were identified by MS. Levels of proteins involved in ROS detoxification and chapero… Show more

“…The oligopeptide samples were analysed by matrix-assisted laser desorption/ionization (MALDI) mass spectrometry (UltraFlex III MALDI ToF/ToF mass spectrometer, Bruker Daltonics, Germany) in MS mode, as per the manufacturer's instructions and exactly as per the calibration and spectral acquisition protocol described earlier (Panda et al 2014). Similarity searches were carried out using MASCOT search engine (Matrix Science, UK) using the NCBI non-redundant database.…”

“…The proteins were applied to 3-10 NL IPG strips, 17 cm (BioRad, India), by cup-loading method, as per the manufacturer's instructions and as detailed earlier (Anaganti et al 2014;Panda et al 2014). Second dimensional resolution was achieved by 14 % SDS-PAGE.…”

“…Due to this complex subcellular membrane organization in cyanobacteria very little is known about the process of extra-cytoplasmic protein sorting. Although cytosolic proteome of this organism has been explored to a large extent [19][20][21][22][23][24][25] there are only few reports [26,27] dealing with membrane proteins and those have employed primarily gel less approach. Employing gel less approach a total of 55 and 18 proteins were identified in the outer membrane fraction of vegetative cells and hetrocysts of Anabaena [26,27].…”

“…Thus, studying the detailed basis of oxidative stress resistance in Anabaena is essential for development of novel biofertilizers, which may be used under adverse conditions. The role played by various proteins in overcoming oxidative stress in the filamentous, heterocystous, nitrogen-fixing, cyanobacterium Anabaena PCC 7120 has garnered remarkable interest in the recent past (Cha et al, 2007;Agrawal et al, 2014;Panda et al, 2014;Banerjee et al, 2015;Tailor and Ballal, 2015).…”
