Mikroelektrophorese an kontinuierlichen Polyacrylamid-Gradienten-gelen, I. Herstellung und Eigenschaften von Gelgradienten in Kapillaren; ihre Anwendung zur Proteinfraktionierung und Molekulargewichtsbestimmung

Abstract

Zusammenfassung: Durch alleinige Ausnutzung der Kapillarattraktion können in Kapillaren mit einem inneren Durchmesser von weniger als l mm kontinuierliche Gelgradienten erzeugt werden. Dafür werden l-, 2-, 5-oder l (^/-Kapillaren zunächst bis zur Hälfte mit einer AmmoniumperoxodisulfatLösung und anschließend mit einer Lösung der restlichen Gelbestandteile gefüllt. Die konzentrierte Gellösung wird dabei kontinuierlich verdünnt wie durch einpolymerisiertes Protein nach Amidoschwarzfärbung und Mikrodensitometrie gezeigt werden kann. Die Mikro-Gradienten-Gele sind zur Fraktionierung komplexer Proteingemische im Molekulargewichtsbereich von l O 4 bis l O 7 geeignet, da die Acrylamidkonzentration von 1,2 bis zu 40% ansteigt. Die Steigung der linearen Gradienten kann in weitem Umfang variiert und somit verschiedenen Trennproblemen angepaßt werden. Die Fraktionierung beruht überwiegend auf dem Molekularsiebeffekt der kontinuierlich geänderten Porenweite. Unter normalen Elektrophoresebedingungen wandert ein definiertes Protein nur so weit im Gel, bis es den seinem Molekulargewicht entsprechenden Platz erreicht hat. Daher können an Mikro-Gradientengelen auch Molekulargewichtsbestimmungen durchgeführt werden.
