Multifunctional Antioxidants for the Treatment of Age-Related Diseases

Abstract: Analogs of N,N-dimethyl-4-(pyrimidin-2-yl)-piperazine-1-sulfonamide possessing either a free radical scavenger group (FRS), chelating groups (CHL), or both (FRS+CHL) have been synthesized. Electrospray ionization mass spectrometry studies indicate that select members of this series bind ions in the relative order of Cu 1+ = Cu 2+ > Fe 2+ = Fe 3+ >Zn 2+ with no binding of Ca 2+ or Mg 2+ observed. In vitro evaluation of these compounds in human lens epithelial, human retinal pigmented epithelial, and human hippo… Show more

“…Indeed, there is a burgeoning literature suggesting strongly that therapies targeted at limiting oxidative stress and inflammation may block early biochemical/cellular alterations in retina even before they are clinically evident. 57,58 Such therapies, therefore, have tremendous potential for slowing/preventing progression of degenerative retinal diseases. Whether FAE may fit this bill, at present, we do not know.…”

“…Since the synthesis of compounds 4 and 8, as described by Jin et al [25], is straight-forward, we believe that the synthetic scheme is amenable to scaled-up GMP production. In the present studies, the study durations versus the amount of synthesized drug on hand limited the doses utilized for each study.…”

“…The use of natural and synthetic antioxidants has shown good potential in reducing ROS and metal ion-induced cell damage [1,141]. Current research efforts, such as that of Kador and co-workers [144] are geared toward the development of multi-functional small drug molecules with chelation and anti-oxidant properties (94, Fig. 25).…”
