Multiple Immune Features-Based Signature for Predicting Recurrence and Survival of Inoperable LA-NSCLC Patients

Abstract: Introduction The immune status of the tumor microenvironment is extremely complex. One single immune feature cannot reflect the integral immune status, and its prognostic value was limited. We postulated that the immune signature based on multiple immuno-features could markedly improve the prediction of post-chemoradiotherapeutic survival in inoperable locally advanced non-small-cell lung cancer (LA-NSCLC) patients. Methods In this study, 100 patients who were diagnosed… Show more

“…Calibration curve determines the consistent degree of fit between actual and predicted values. The reference line of the calibration curve is 45 degree and indicates perfect calibration [ 11 ]. As shown in Figure 5 , the fitness of TZI (left panel), which is closer to the 45-degree diagonal, is better than that of CTZI (right panel).…”

“…Because the DNA of tumor cells that shed stool cannot contain a single gene, the sensitivity and specificity of single gene methylation has not reached a very satisfactory level. Combined multitarget detection has become a common method to improve detection sensitivity ( 25 , 38 41 ). A previous study reported that the sensitivity of the stool DNA methylation test using single and multiple genes was 48% and 77.8%, and the specificity was 97% and 92.7%, respectively ( 42 ).…”

“…An emerging study reported the immune-excluded phenotype exits a su cient amount of immune cell but could not completely elicit anti-tumor immune responses since immune cells remain in the stroma surrounding the tumor cell nest instead of penetrating its substance (43,44). The TIIC cluster C was de ned as the immune-desert phenotype characterized by immune suppression, which means the immune tolerance and escape, decreasing in ltration of lymphocytes and lacking activated priming T cells (45). Meanwhile, we found that TIIC cluster C had a highest stromal score, which is widely considered to be positively associated with the TAMs and resting DCs, the results indicated that a humoral immune response in immune-desert phenotype (46, 47).…”
