New Insights on Saccus vasculosus Evolution: A Developmental and Immunohistochemical Study in Elasmobranchs

Abstract: The saccus vasculosus (SV) is a circumventricular organ of the hypothalamus of many jawed fishes whose functions have not yet been clarified. It is a vascularized neuroepithelium that consists of coronet cells, cerebrospinal fluid-contacting (CSF-c) neurons and supporting cells. To assess the organization, development and evolution of the SV, the expression of glial fibrillary acidic protein (GFAP) and the neuronal markers γ-aminobutyric acid (GABA), glutamic acid decarboxylase (GAD; the GABA synthesizing enzy… Show more

“…These results suggest that coronet cells in the SV contain entire photoperiodic signalling pathway from light input (photoreceptor) to neuroendocrine output (TSHb and DIO2). The coronet cells have traditionally been considered to be responsible for the function of the SV [14][15][16] , and our results are consistent with this notion. As the circadian clock is involved in photoperiodic time measurement, we also examined and confirmed expression of circadian clock genes in the SV ( Supplementary Fig.…”

“…1a-e). The SV is a specialised circumventricular organ found in the caudal hypothalamus of many jawed fish, but its physiological function remains unclear [14][15][16] . Biological activity of TSH requires heterodimerization of the TSH b-subunit and a common glycoprotein a-subunit.…”
