Production and Characterization of a Monoclonal Antibody Specific for Salmonella O19-Antigen

Abstract: A monoclonal antibody, SBY1 (IgM, kappa), against the Salmonella O-antigen was generated by using the myeloma cell line Sp2/O-Ag14 as a fusion partner with spleen cells from BALB/c mice immunized with S. senftenberg 963 K. SBY1 was characterized by the slide agglutination and absorption test. SBY1 was believed to show the specificity to O1-, O3- or O19-antigens of Salmonella because S. Senftenberg 963 K (O1, 3, 19) was used as the antigen for immunization. The slide agglutination test with the Salmonella serov… Show more

“…Production of O:5 and O:8 specific MAbs have been reported [21,23,24,25] but cross‐reactions with other Salmonella serogroups were not observed. Common antigenic determinants of group B and group C 2 –C 3 Salmonella strains also have not been described by the classical serology [18].…”

“…Other data also point out that the acetylation may a¡ect the immunological properties of the O-antigen and may alter polyclonal antibody response after oral immunization with S. typhimurium, which correlates with protection [22]. “…A few attempts to generate MAbs specific for Salmonella Typhimurium and other serogroup B salmonellae have been made, however, not one against OMP antigens. MAbs were obtained against LPS antigens from these bacteria but all presented cross-reactivity with strains from other serogroups (Mitov et al, 2003;Tsang et al, 1991;Yamaura, Uchiyama, & Terkado, 1992).…”

“…Outbreaks caused by this pathogenic strain have been reported in broiler chickens and pigeons (9,22). Since S. typhimurium infections of animals play an important role in zoonosis, it has been suggested that the serological differentiation of the types of S. typhimurium by specific antibodies could be an effective approach for the identification of the source of infection by this pathogen (29).…”

“…Although the reported MAb was more specific than polyclonal antibodies, it exhibited some cross-reaction with other Salmonella serotypes. Recently, MAbs against heat-attenuated S. typhimurium reactive with the LPS O-5 antigen have been developed (29). The latter antibodies showed various reactions with the serovars that may carry the O5 antigen.…”
