Protective Effect of Sulfated Polysaccharides from Celluclast-Assisted Extract of Hizikia fusiforme Against Ultraviolet B-Induced Skin Damage by Regulating NF-κB, AP-1, and MAPKs Signaling Pathways In Vitro in Human Dermal Fibroblasts

Abstract: Our previous study evaluated the antioxidant activities of sulfated polysaccharides from Celluclast-assisted extract of Hizikia fusiforme (HFPS) in vitro in Vero cells and in vivo in zebrafish. The results showed that HFPS possesses strong antioxidant activity and suggested the potential photo-protective activities of HFPS. Hence, in the present study, we investigated the protective effects of HFPS against ultraviolet (UV) B-induced skin damage in vitro in human dermal fibroblasts (HDF cells). The results indi… Show more

“…Inhibitory effect on collagenase was performed by modified Wang's method [27]. To measure the collagenase activity, 100 µL of 200 units/mL collagenase and 100 µL PNPs (5000, 2500, 1250 µg/mL) were mixed together and incubated at 37 • C for 15 min, weighed at 1 mg of azo dye-impregnated collagen substrate and mixed with 800 µL 0.1 M Tris-HCl buffer (pH 7.0), then added together at 43 • C for 1 h under shaking conditions.…”

“…Natural products, such as polyphenols, alkaloids, terpenoids, organosulfur compound, phytosterols, and bioactive compounds obtained from land-based and marine resources, were shown to exhibit anti-obesity effects through the regulation of obesity-related risk factors [23][24][25][26][27][28]. Marine seaweeds are a potential rich source of bioactive metabolites, including polyphenols, polysaccharides, and peptides, that exhibit antioxidant, anticancer, antidiabetic, anti-inflammation, anti-ultraviolet, and anti-obesity activities [29][30][31][32][33][34][35][36][37]. Many researchers have reported that marine seaweed metabolites, such as alginates, fucoidans, and phlorotannins, have potential anti-obesity effects in in vivo animal studies [38].…”

“…It has been used as a food ingredient and herbal medicine for hundreds of years (Li et al 2006). H. fusiforme contains a particular high amount of polysaccharides that possess various bioactivities (Wang et al 2018a). In a previous study, we suggested that sulfated polysaccharides isolated from a Celluclast-assisted extract of H. fusiforme (HFPS) possess strong antioxidant activities in vitro and in vivo (Wang et al 2018b).…”
