Simultaneous Determination of Tetracycline, Macrolide and Sulfonamide Antibiotics in Soils Using Accelerated Solvent Extraction Followed by Solid-Phase Extraction and High Performance Liquid Chromatography Tandem Mass Spectrometry

Abstract: This research developed and optimized the methods for simultaneous determination of tetracycline (TCs), macrolide (MLs) and sulfonamide (SAs) antibiotics in soils using accelerated solvent extraction (ASE)- solid-phase extraction (SPE)-high performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS). Methanol-citric acid (pH=4.7) was used as extraction solvent, diatomite (washed by EDTA) was used as dispersing agent. Firstly, soil was extracted by ASE with the parameter conditions: pressure 1500 ps… Show more

“…Methanol, acetone, acetonitrile, dichloromethane, and ethyl acetate are solvents commonly used for the extraction of various pesticide and veterinary drugs, of which methanol is the solvent most used to dissolve and extract polar and nonpolar chemicals. In many previous studies, methanol has been widely used in the extraction of veterinary drugs, such as tetracyclines, macrolides, and sulfonamide antibiotics [ 14 , 26 , 27 , 28 , 29 , 30 ]. Extraction conditions are mainly associated with the physical and chemical features of the target compounds as well as the features of the solvents.…”

“…A wide variety of methods have been employed to determine antibiotic compounds from soil and water samples. The methods utilized most frequently for soil extraction are liquid-solid extraction (LSE) [11,12], ultrasonic assisted extraction (UAE) [13][14][15], accelerated solvent extraction (ASE) [16][17][18], and pressurized liquid extraction (PLE) [19,20]. Sample clean up and concentration of water samples and soil extracts typically occurs via solid phase extraction (SPE) [18,21,22].…”

“…Sample pretreatment is an important process for enhancing the sensitivity and accuracy of analytical methods. In previous studies, various methods have been proposed, such as solid-phase extraction, 18–21 QuEChERS (Quick, Easy, Cheap, Effective, Rugged, and Safe), 22 and matrix solid-phase dispersion extraction (MSPD). 23 Among them, QuEChERS was considered as a preferred procedure because of its simplicity, rapidity, and environmental friendliness.…”
