Site‐specific cleavage of acetoacetyl‐CoA synthetase by legumain

Abstract: Acetoacetyl-CoA synthetase (AACS) is a ketone body-utilizing enzyme and is responsible for the synthesis of cholesterol and fatty acids. We have previously shown that AACS is cleaved by legumain, a lysosomal asparaginyl endopeptidase. In this study, we attempted to determine the cleavage site of AACS. Mutagenesis analysis of AACS revealed that Asn547 is the specific cleavage site of AACS in mouse livers. The cleaved form of AACS (1-547) lost the ability to convert acetoacetate to acetoacetyl-CoA. Moreover, hyd… Show more

“…AACS has the ability to use ketone bodies for the creation of cholesterol and fatty acids, converting acetoacetate into its CoA ester to generate acetoacetyl-CoA in the cytosol [108]. This process is facilitated by HMGCS, which transforms acetoacetyl-CoA into 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) [109,110]. Subsequently, HMGCRa is able to facilitate the transformation of HMG-CoA into mevalonic acid in order to generate cholesterol [111].…”

“…Whereas, for the moment, little knowledge of ACSF family in cancer has been reported until now. Hasegawa et al [ 15 , 16 ] demonstrate that the limited proteolysis of AACS can encode the ketone body-utilizing enzyme, and is adjusted transcriptionally by SREBP-2. Meanwhile, the ketone body metabolism regulated by AACS plays a significant role in cholesterol metabolism.…”

“…After 2 days of transfection, the cells were harvested in cell lysis buffer (10 mM Tris-HCl pH7.4, 1% NP40, 150 mM NaCl, and 0.5 mM EDTA). AACS activity in 50 μg of protein extracts was assayed by measuring acetyl-CoA formation according to a method previously reported by Hasegawa et al 14…”
