Background Cancer stem cells (CSCs) are crucial to the malignant behaviour and poor prognosis of pancreatic ductal adenocarcinoma (PDAC). In recent years, CSC biology has been widely studied, but practical prognostic signatures based on CSC-related genes have not been established or reported in PDAC. Methods A signature was developed and validated in seven independent PDAC datasets. The MTAB-6134 cohort was used as the training set, while one local Chinese cohort and five other public cohorts were used for external validation. CSC-related genes with credible prognostic roles were selected to form the signature, and their predictive performance was evaluated by Kaplan–Meier survival, receiver operating characteristic (ROC), and calibration curves. Correlation analysis was employed to clarify the potential biological characteristics of the gene signature. Results A robust signature comprising DCBLD2, GSDMD, PMAIP1, and PLOD2 was developed. It classified patients into high-risk and low-risk groups. High-risk patients had significantly shorter overall survival (OS) and disease-free survival (DFS) than low-risk patients. Calibration curves and Cox regression analysis demonstrated powerful predictive performance. ROC curves showed the better survival prediction by this model than other models. Functional analysis revealed a positive association between risk score and CSC markers. These results had cross-dataset compatibility. Impact This signature could help further improve the current TNM staging system and provide data for the development of novel personalized therapeutic strategies in the future.

Sendo conceituado o sistema nervoso central (SNC) como um sistema compartimentalizado que guarda relativa autonomia em relação ao restante da economia do corpo, os processos imunológicos mantêm nele certas particularidades. Por um lado, a barreira hêmato-encefálica restringe a entrada de anticorpos e de antígenos sistêmicos. Por outro lado, o SNC e as estruturas continentes do líquido cefalorraqueano (LCR) carecem de estruturas linfóides organizadas e têm poucos linfócitos participando de sua população celular em condições normais.Em função desses fatores e baseando-se na homologia existente entre os linfócitos do LCR e os do restante da economia, é necessário adotar técnicas especiais para o estudo de subpopulações linfocitárias do LCR. Constitui o objeto deste estudo descrever a técnica padronizada para o estudo dessas subpopulações celulares do LCR, a qual permite reprodutibilidade segura mesmo quando amostras oligocelulares, como as de LCR normal são consideradas. MÉTODOPreparo da amostra -Imediatamente após a colheita do LCR, em amostra de 10 ml é feita a contagem global de células em câmara tipo Fuchs-Rosenthal e a análise citomorfológica em preparados obtidos por sedimentação gravitacional acelerada segundo Suta e coradas, posteriormente, segundo Leishman 14.

For cerebrospinal fluid (CSF) samples from 50 subjects previously selected to represent suitably a normal control group and for a methodology that may be actually used for CSF samples even with normal cell count, it is possible to characterize lymphocyte subpopulations with normal statistical distribution and low dispersion of results for all lines considered. Data (mean +/- standard deviation) here estimated were (%): for B-lymphocyte subpopulation 7.0 +/- 1.44; for T lymphocyte subpopulation 76.0 +/- 2.92; for T-active lymphocytes 53.7 +/- 3.26; for T-avid lymphocytes 19.2 +/- 4.32 when all T subpopulation is considered, and 11.1 +/- 2.04 when only T-active subpopulation is studied; for T-sensitized or actually activated 22.3 +/- 3.95. These data may suggest that lymphocytes actually related to immunological activation will be properly represented by the difference between T-lymphocytes and its T-active line in each case. In every samples T and T-active lymphocytes are different subpopulations when surface receptor density is considered, by evidence of avidity for sheep erythrocytes. It is also possible that killer lymphocytes, or antibody mediated cytotoxic cells, may be related to modified lymphoid cells identified in the CSF normal cytomorphological profile.
