Untangling cell tracks: Quantifying cell migration by time lapse image data analysis

Abstract: Automated microscopy has given researchers access to great amounts of live cell imaging data from in vitro and in vivo experiments. Much focus has been put on extracting cell tracks from such data using a plethora of segmentation and tracking algorithms, but further analysis is normally required to draw biologically relevant conclusions. Such relevant conclusions may be whether the migration is directed or not, whether the population has homogeneous or heterogeneous migration patterns. This review focuses on t… Show more

“…Images of motile ECs are captured with a 5 min time interval over 4 h. Images were then processed with DIAS software (Solltech). A recent review on tracking algorithms offers a wide and comprehensive selection of the available tools to analyze cell motility [49]. Generally, data are displayed as a centroid plot showing the location of the geometrical center of the cell as a function of time.…”

“…It is well accepted that speed and meandering index can be used for determining persistence, also defined as the period by which a cell moves in the same direction. 45 The 3 J/cm 2 laser-treated group demonstrated considerably higher net distances toward wound and higher meandering index value. These macrophages demonstrated a preferential wound migration bias with less ''stopping'' and lower static ratios than the untreated controls, indicating more persistent or efficient wound-oriented tracks.…”

“…TLM is the technique of capturing the sequence of microscopic images at regular intervals. TLM allows scientists to observe cellular dynamics and behavior of the population of living cells as well as of the single living cell within the population [2,3]. Live cell imaging and the first nonsophisticated TLM techniques were pioneered at the very beginning of the twentieth century [4].…”
