The melanocortin system is composed by the agonists adrenocorticotropic hormone and α, β and γ-melanocyte-stimulating hormone, and two naturally occurring antagonists, agouti and agouti-related protein. These ligands act by interaction with a family of five melanocortin receptors (MCRs), assisted by MCRs accessory proteins (MRAPs). MCRs stimulation activates different signaling pathways that mediate a diverse array of physiological processes, including pigmentation, energy metabolism, inflammation and exocrine secretion. This review focuses on the regulatory mechanisms of MCRs signaling, highlighting the differences among the five receptors. MCRs signal through G-dependent and independent mechanisms and their functional coupling to agonists at the cell surface is regulated by interacting proteins, namely MRAPs and β-arrestins. The knowledge of the distinct modulation pattern of MCRs signaling and function may be helpful for the future design of novel drugs able to combine specificity, safety and effectiveness in the course of their therapeutic use.

A c c e p t e d M a n u s c r i p t SummaryMelanocortin 5 receptor (MC5R) is a G-protein coupled receptor (GPCR) with high affinity for α-MSH. To unravel some of the downstream cell-signaling pathways activated by this receptor, HEK293 cells were transiently and stably transfected with a vector encoding green fluorescent protein (GFP)-tagged MC5R. In these cells the receptor was correctly addressed to the cell surface and was functional, as shown by the MC5R-induced formation of intracellular cAMP. In fact, the MC5R agonist α-MSH induced a 10 or 16 fold increase (transient or stable cells, respectively) above the cAMP levels found in unstimulated cells. Moreover, in cells stably expressing MC5R-GFP, α-MSH promoted ERK1/2 phosphorylation in a dose-dependent manner (EC50=7.3nM) with the maximal effect occurring after 5min of agonist incubation. The signaling pathway conveyed through ERK1/2 is not linked to cAMP, since the phosphorylation of these kinases is unchanged by the inhibition of adenylyl cyclase. Also, ERK1/2 activation is not significantly affected by PKA, PKC and Akt/PKB specific inhibitors. However, α-MSH-induced ERK1/2 activation is abolished by the PI3K inhibitors wortmannin and LY294002. Altogether, these findings demonstrate that MC5R signals through a PI3K-regulated Akt-independent pathway leading to downstream activation of ERK1/2. The involvement of these MAPK suggests that MC5R could be implicated in cellular proliferation or differentiation mechanisms.
Epilepsy is the most common neurological disorder worldwide. Studies have shown that recurrent seizures may increase the concentration of reactive oxygen species, which can lead to oxidative stress and neuronal damage. These seizures result in substantial deleterious effects on an individual's health. Organic and conventional grape juices are rich in polyphenols, compounds with important antioxidant activity. However, these juices could have differences in their polyphenol content. The aim of this study was to investigate the neuroprotective and anticonvulsant effects of organic and conventional grape juice treatments in Wistar rats against pentylenetetrazole (a convulsant drug)-induced damage. In addition, we evaluated potential behavioral changes in rats treated with the juices and the polyphenolic profile of those samples. Animals (n=16 in each group) received treatment with saline, organic or conventional grape juice for 17 days. On the eighteenth day, behavioral changes were evaluated by an open field test. Afterwards, half of the rats from each group received pentylenetetrazole and were observed for 30 min to evaluate possible seizure characteristics. The animals were subsequently killed by decapitation and their hippocampus, cerebellum and cerebral cortex tissues were isolated. The results of this study showed that neither organic nor conventional grape juice altered the behavior parameters, and no statistical differences were observed in the seizure characteristics of the groups. Nevertheless, both juice types were able to protect from lipid and protein oxidative damage, decrease nitric oxide content and increase enzymatic (superoxide dismutase and catalase) and non-enzymatic (sulfhydryl protein) antioxidant defenses in brain tissues following pentylenetetrazole-induced seizures. In general, organic juice showed superior results in each test, probably due to its higher polyphenol content relative to conventional juice. These results indicate that grape juices can provide further insight into natural neuroprotective compounds and may lead to the development of new therapeutic strategies for epileptic patients.
As verminoses ainda são um sério problema de saúde pública no Brasil. Elas são transmitidas facilmente a crianças em idade escolar em consequência de seu sistema imunológico imaturo e da precariedade na manutenção de hábitos saudáveis de higiene pessoal. O objetivo deste trabalho foi avaliar a prevalência de parasitoses e o perfil socioeconômico da população escolar de Flores da Cunha-RS. Foram coletadas amostras fecais dos alunos para posterior análise por sedimentação espontânea e aplicados questionários acerca das parasitoses e da situação socioeconômica. Das 341 amostras avaliadas, 10,0% foram positivas, sendo observada a presença de cistos de Endolimax nana (55,9%), de Entamoeba coli (26,5%), de Iodamoeba butschlii (5,9%), de Giardia lamblia (2,9% ) e ovos de Ascaris lumbricoides (2,9%), além de indivíduos (5,9%) com parasitose múltipla (Entamoeba coli e Endolimax nana). A maioria dos responsáveis afirma buscar informações sobre parasitoses com profissionais da saúde (49,7%), mesmo assim 49,9% não acham suficientes as informações disponíveis. Em relação ao saneamento básico, 57,2% declaram possuir coleta e tratamento de esgoto, 67,2% têm boa captação da chuva, o que evita acúmulos nas proximidades, e 47,2% fazem algum tipo de controle de pragas. Entre os escolares, 79% nunca fizeram exame parasitológico de fezes ou foram diagnosticados com parasitose, mesmo assim 85,9% afirmaram ter usado antiparasitários. A baixa prevalência de parasitoses encontrada no presente estudo é justificada, sobretudo, pelo constante uso de antiparasitários. Desse modo, é importante melhorar a divulgação de orientações sobre a prevenção de parasitoses e o perigo da automedicação.
Oxidative damages in hepatocytes may be caused by epilepsy and/or anticonvulsant drugs. Epilepsy is one of the most common neurological disorders, characterized by recurrent seizures, which may increase the content of reactive oxygen species. Organic and conventional grape juices are rich in polyphenols, compounds with important antioxidant activity. It is hypothesized that organic and conventional purple grape juices may have protective effect against oxidative damage induced by pentylenetetrazole (PTZ) (a standard convulsant drug) in the liver and serum of Wistar rats. Animals (n = 16 in each group) received, by gavage, saline, organic grape juice or conventional grape juice (10 μL/g of body weight) for 17 days. Subsequently, half of the rats in each group received PTZ (60 mg/kg). After 30 minutes, the animals were euthanized by decapitation. Liver and blood samples were isolated to evaluate oxidative parameters (lipid and protein oxidation, nitric oxide metabolite content, antioxidant defenses, and protein sulfhydryl content). The results of this study showed that although organic juice contains higher polyphenol content than conventional juice, both juices conferred protection against lipid and protein oxidative damage and limited the increase in PTZ-induced nitric oxide metabolite content in the liver and serum. In addition, both juices inhibited the PTZ-induced reduction in enzymatic antioxidant defenses (superoxide dismutase and catalase activities) and sulfhydryl protein content in the liver and serum. In summary, both organic and conventional grape juices were able to reduce oxidative damage induced by PTZ in the liver and serum of Wistar rats.
