Investigating the function of combining induced rat monocytes-derived bone marrow-haemopoietic stem cell (rat BM-HSCs) with LPS and rat bone marrow-mesenchymal stem cell (rat BM-MSCs) wasto analyze the acceleration of homing process mechanism in injured pancreas. Mononucleated stem cells were isolated from aspirated whole rat BM using ficoll and cultured in α-MEM complete growth medium in 10 cm petridish. After two days, adherent cells after washing twice in petridish were added α-MEM growth medium and then mesenchymal cells were characterized using CD105 marker in third passage and labeled PKH26. Then haemopoietic stem cells (HSCs) were isolated with magnetic beads CD34+ and differentiated in vitro, and then induced monocytes with LPS. Animal experiment used 28 male Wistar rats, and divided them into 4 groups. After transplantation combined, both cells between monocyte derived HSc (mHSCs) and rat BM-MSC were analyzed expression of pair box gen 4 (Pax4), pancreatic and duodenal homeobox (Pdx1), C-peptide using immunohistochemistry, then secretion of insulin and C-peptide analyzed using in-direct ELISA. Results showed that the expressions of Pax4, Pdx1, C-peptide found in the surface membrane cell F. A. Rantam et al. 334 of pancreatic cell, and secreted C-peptide and insulin were shown significant (P < 0.05) in transplanted group 2, 3 and 4, but in group 3 were transplanted with combined cells more dominant than non-combined cells. Conclusions suggested that combining of induced monocytes-derived HSCs and rat BM-MSCs has accelerated homing MSCs into injured pancreatic tissue.
Human Adipose Derived Mesenchymal Stem Cells (HADMSCs) and Human Hair Follicle Derived Mesenchymal Stem Cells (HHFDMSCs) have attracted great interest because of their multilineage differentiation potential, selfrenewal properties, and their possible use of cell and gene therapies. This present study to investigate the neurogenic differentiation ability of hADMSCs and hHFDMSCs induced by Epidermal Growth Factors (EGF), Fibroblast Growth Factor (FGF), Platelet Derived Growth Factor (PDGF) and Forskolin. This study was true experimental with longitudinal study design. The sample size determined with minimal sample size formula and it was randomly chosen. These studies employed an in vitro design for the expansion and proliferation of Mesenchymal Stem Cells (MSCs) and examined the heterogeneity of these cells using the markers CD105, CD90, OCT4, and SOX2. MSCs from adipose tissue and hair follicles were induced with EGF, FGF, PDGF and Forskolin to differentiate and generate neurons. The capacity of MSCs to generate neurons were verified using glial fibrillary acidic protein, nestin, and β-tubulin III. The expression of neural markers and morphological changes in Mesenchymal stem cells from hADMSCs and hHFDMSCs were confirmed. hADMSCs and hHFDMSCs share a similar capacity to differentiate and generate neurons, which is beneficial for the development of neuronal restoration for future therapies for patients suffering from neurological diseases.
Indonesia mengikuti target eliminasi global yaitu Eliminasi TB 2035 dan Indonesia bebas TB 2050. Tantangan dalam program pengendalian Tuberkulosis di Indonesia adalah banyaknya kasus tuberkulosis yang tidak terdeteksi maupun kasus yang sudah terdeteksi namun belum terlaporkan, sehingga perlu dilakukan perubahan pendekatan penemuan kasus, dari secara pasif menjadi intensif dan aktif. Rumitnya pengisian form TB oleh dokter swasta, membuat rendahnya notifikasi pasien TB. Maka dibuat aplikasi berbasis smartphone yaitu Aplikasi Wajib Notifikasi TB (WiFi) TB yang lebih sederhana dan mudah digunakan. Penelitian ini bertujuan untuk menggambarkan pelaksanaan wajib noifikasi (WiFi) TB di Dinas Kesehatan Kabupaten Jember. Penelitian ini menggunakan jenis penelitian deskriptif. Sampel penelitian adalah 2 petugas TB. Penelitian ini dianalisis menggunakan analisis deskriptif. Hasil dari penelitian ini menunjukkan bahwa gambaran dari aspek input, proses, dan output dalam pelaksanaan wajib notifikasi (WiFi) TB
Objective: The purpose of this review literature is to look at the mechanism of mesenchymal stem cell secretome in the regeneration of periodontal tissue. Methods: This literature review was collected from several journals and research that has been done, where the application of Mesenchymal stem cells (MSCs) promises a favourable result for cellbased therapy in inflammatory diseases, immune system disorders, regeneration, and tissue repair. Results: MSCs possess a potent immunomodulatory function, paracrine effects, and the ability to differentiate into different cells. Not only mesenchymal stem cells can replace damaged tissue or cells but also secrete secretome namely bioactive ingredients such as cytokines, chemokines, growth factors, pro-inflammatory, and anti-inflammatory which play an essential role in regulating the host immune system. Periodontal disease is an inflammatory disease that occurs in the teeth supporting structures. Lately, many therapies for periodontal disease utilized MSCs from gingival tissue and periodontal ligaments. Secretome activates endogenous MSCs to improve the immune system in periodontal disease that might create a better microenvironment of periodontal tissue. Conclusion:The secretome of MSCs has attracted a lot of attention and has the potential to repair and regenerate tissues including periodontal tissue.
Side effects of the use of contraceptives have always been a problem complained by family planning acceptors. The side effects of family planning such as irregular menstruation, weight gain, nausea, dizziness, and vaginal discharge. This research was conducted in July-August 2019 to determine the factors causing the selection of contraceptives by family planning acceptors at the Sumbersari Public Health Center in Lumajang Regency. Researchers used a cross-sectional design. The sample were 98 family planning acceptors at the Sumbersari Public Health Center, Lumajang Regency, selected using cluster random sampling technique. Researchers used statistical regression tests to data analyze process. Statistical test results of factors that had a positive influence on the selection of contraceptive devices for family planning acceptors in the working area of Sumbersari Public Health Center were the number of children, knowledge of family planning, availability of tools, attitudes and husband's support; counseling performance in the majority of acceptors was good, and the majority of acceptors had a negative family planning history.
