Plant growth retardant (PGR) refers to organics that can inhibit the cell division of plant stem tip sub-apical meristem cells or primordial meristem cell. They are widely used in the cultivation of rhizomatous functional plants; such as Codonopsis Radix, that is a famous Chinese traditional herb. However, it is still unclear whether PGR affects the medicinal quality of C. Radix. In the present study, amino acid analyses, targeted and non-targeted analyses by ultra-performance liquid chromatography combined with time-of-flight mass spectrometry (UPLC-TOF-MS) and gas chromatography-MS were used to analyze and compare the composition of untreated C. Radix and C. Radix treated with PGR. The contents of two key bioactive compounds, lobetyolin and atractylenolide III, were not affected by PGR treatment. The amounts of polysaccharides and some internal volatiles were significantly decreased by PGR treatment; while the free amino acids content was generally increased. Fifteen metabolites whose abundance were affected by PGR treatment were identified by UPLC-TOF-MS. Five of the up-regulated compounds have been reported to show immune activity, which might contribute to the healing efficacy (“buqi”) of C. Radix. The results of this study showed that treatment of C. Radix with PGR during cultivation has economic benefits and affected some main bioactive compounds in C. Radix.
Adsorption and desorption mechanisms of methylene blue (MB) removal with iron-oxide coated porous ce-ramics filter (IOCPCF) were investigated in batch and column mode. The results revealed that MB removal mechanisms included physical adsorption and chemical adsorption, of which chemical adsorption by surface ligand complex reaction played a dominant role after infrared spectrum analysis. Recycling agents were se-lected from dilute nitric acid (pH=3), sodium hydroxide solution (pH=12) and distilled water. Among three agents, dilute metric acid (pH=3) was the best recycling agent. Regeneration rate of IOCPCF arrived at 82.56% at batch adsorption and regeneration was finished in 75min at column adsorption. Adsorp-tion-desorption cycles of IOCPCF after batch and column adsorption were four and three times, respectively. Further, compared with fresh IOCPCF, MB removal rate with these desorbed IOCPCF adsorption only slightly decreased, which suggested that IOCPCF should be used repeatedly
Hibiscus mutabilis (cotton rose) is a deciduous shrub or small tree of the Malvaceae family. Here, we report a chromosome-scale assembly of the H. mutabilis genome based on a combination of single-molecule sequencing and Hi-C technology. We obtained an optimized assembly of 2.68 Gb with a scaffold N50 length of 54.7 Mb. An integrated strategy of homology-based, de novo, and transcriptome-based gene predictions identified 118,222 protein-coding genes. Repetitive DNA sequences made up 58.55% of the genome, and LTR retrotransposons were the most common repetitive sequence type, accounting for 53.15% of the genome. Through the use of Hi-C data, we constructed a chromosome-scale assembly in which Nanopore scaffolds were assembled into 46 pseudomolecule sequences. We identified important genes involved in anthocyanin biosynthesis and documented copy number variation in floral regulators. Phylogenetic analysis indicated that H. mutabilis was closely related to H. syriacus, from which it diverged approximately 15.3 million years ago. The availability of cotton rose genome data increases our understanding of the species’ genetic evolution and will support further biological research and breeding in cotton rose, as well as other Malvaceae species.
Fluorescent carbon nanoparticles (f-CNPs) were successfully grafted with polystyrene by employing a "grafting from" method via reversible addition-fragmentation chain transfer polymerization (RAFT). The detailed polymerization kinetic studies showed that the grafting polystyrene polymerization from the f-CNPs surface via RAFT was living-controlled. The synthesized f-CNP-g-PSt nanocomposites with different grating lengths via RAFT could fluoresce like f-CNPs. At the same time, the synthesized f-CNP-g-PSt nanocomposites could be easily processed into fibers by electrospinning. The fibers could still fluoresce at the appropriate excitation wavelength. The f-CNP-g-PSt nanocomposites synthesized via RAFT feature good processability may be applied as luminescent plastic.
