Luka bakar adalah kerusakan jaringan pada kulit akibat terpajan panas tinggi, bahan kimiawi maupun arus listrik. Salah satu hewan yang disembelih di Rumah Potong Hewan (RPH) adalah sapi. Seekor sapi dapat menghasilkan limbah darah kurang lebih sebanyak 28 liter. Komponen darah terdiri dari plasma, sel darah merah, sel darah putih dan platelet. Platelet mengandung growth factor. Platelet Rich Plasma (PRP) atau plasma yang kaya akan platelet terbukti dapat mempercepat penyembuhan tulang dan jaringan lunak. Pembuatan PRP dari darah sapi dilakukan dengan metode sentrifuge. Hewan coba yang dipakai pada penelitian ini adalah tikus putih sebanyak 10 ekor. Besi panas berbentuk bulat dipanaskan dengan api, lalu ditempelkan pada kulit. Kelompok perlakuan diberi PRP dengan konsentrasi 10%, 20% dan 30% dengan salep vaselin album, kelompok kontrol negatif yang tidak diberi perlakuan dan kelompok kontrol positif yang diberi obat yang terbuat dari ekstrak plasenta sapi 10%+neomycin sulfate 0,5%. Parameter yang dipakai untuk mengukur tingkat kesembuhan luka adalah waktu kesembuhan dan pemeriksaan histopatologi. Pada hasil pengukuran waktu kesembuhan dianalisis dengan metode statistik One-way Analsisis of variance (Anova) tingkat signifikansi 95% dengan Kruskal-wallis, kesembuhan luka terdapat perbedaan signifikan antara PRP konsentrasi 10%, 20%, 30%, dan kontrol positif dengan kontrol negatif. Konsentrasi PRP yang optimal adalah 20%. Analisa tipe jaringan yang terbentuk dilakukan dengan pembuatan preparat histopatologi, hasil menunjukkan bahwa luka pada kelompok perlakuan dan kontrol positif tampak sembuh, sedangkan kelompok kontrol negatif epitelisasi belum tertutup jelas. PRP merupakan obat penyembuh luka bakar yang efektif sebagai pengganti obat komersil yang sudah tersedia.
Background and Aim: Enrofloxacin and tylosin can be combined into an antibiotic formulation which is expected to have a broader range of antibacterial activity against various infections in broilers. Validation method analysis of the levels of these two active compounds needs to be done for future use in pharmacokinetic or residual studies. The present study aims to determine a suitable validation method of isocratic high-performance liquid chromatography (HPLC) to measure the concentration of antibiotic combinations in the broiler liver, kidney, and muscles. Materials and Methods: The combination of enrofloxacin and tylosin in the liver, kidney, and muscle was validated by HPLC method to find the procedures, processes, equipment, and systems used, consistently provides the appropriate results. The chromatography system consisted of an Octadecyl-silica column of 5 μm in diameter and 150 mm in length with a mobile phase of a mixture of 0.05 M monobasic sodium phosphate (pH 2.5) and acetonitrile (65:35 v/v). The solution was detected at a wavelength of 280 nm, 30°C, a flow rate of 1 mL/min, and an injection volume of 20 μL. The combination antibiotics powder was produced from PT Tekad Mandiri Citra, Bandung, Indonesia, and broiler tissues obtained from day-old chick broilers maintained for 30 days with free antibiotic feed. Results: Validation of a combination solution of enrofloxacin and tylosin shows the linearity values of enrofloxacin and tylosin in the liver, kidney, and muscles as r2=0.9988, r2=0.9999, r2=0.9997, r2=0.9989, r2=0.9978, and r2=0.9962. The accuracy and precision values of enrofloxacin in the liver, kidney, and muscles were 5.53, 6.23, and 6.93, respectively. The values of accuracy and precision of tylosin in the liver, kidney, and muscles were 10.43, 4.63, and 7.16%, respectively. The retention times for enrofloxacin and tylosin were 1.945-2.000 min and 4.175-4.342 min. The limit of detection (LOD) and limit of quantity (LOQ) values for enrofloxacin were 3.03 and 10.1 μg/g, respectively. In contrast, the LOD and LOQ values for tylosin were 9.05 and 30.17 μg/g, respectively. Conclusion: The value of linearity, accuracy, precision, specificity, and sensitivity of the combined solution of enrofloxacin and tylosin showed promising results.
Number of patients with diabetes mellitus are very large, according to International Diabetes Federation (IDF) in 2013 reaches 382 billion people between the ages of 40-59 years. Diabetic has been characterized as a chronic wounds. The animal blood waste from a slaugther house is still untapped, containing PRP (Platelet Rich Plasma) that can accelerates heal of bone and soft tissue. PRP (Platelet Rich Plasma) was made from the cattle blood waste using centrifugation. The purpose of the study is to prove that the Prima cream can heal the wounds in diabetic patients. Animals used in this study is 20 Wistar rats. Induction of diabetes was using streptozootocin 65 mg/kg and nicotinamide 235 mg/kg. The animal were divided into 4 groups: non-diabetes given placebo (ND-), diabetes given placebo (DM-), diabetic rat given commercial drug were containing chloramphenicol base 20 mg + prednisolone 2.5 mg (DM+), and diabetes given cream PRP at 20% concentration (DM PRP). Parameters used to measure the rate of wound healing is wound size and histopathologic examination. The data were analyzed using Anova One Way at 95% significant rate. PRP DM group had a wound healing faster than ND- group, DM- and DM+. Based on histopathologic examination, such as epitelialization, basal membrane, fibroblasts, collagen tissue, and presence of inflammatory cells, the best curing reaction was showed in DM PRP. PRP 20% cream produces from cow blood waste can accelerate wound healing in diabetes mellitus.
