The effect of sleep restriction and psychological load on the diurnal metabolic changes in tryptamine-related compounds in human urine

Abstract: Objective The effect of a severely stressful situation (sleep restriction and psychological load) on the diurnal changes in novel tryptamine-related compounds (hydroxydiacetyltryptamine, sulphatoxymelatonin, and dihydromelatonin) was evaluated in human subjects for 16 days. Methods The subjects were allowed to sleep for 5 h on days three through 12 and for 8 h on the other days. On days three through 12, the subjects were asked to perform a psychological task. The first two and the last 4 days were viewed as c… Show more

“…Urinary 24-h melatonin concentrations decreased by more than 50% when subjects slept five hours versus eight hours, while being continuously exposed to white light (700 lux) during wake hours. 161 Diminished levels of melatonin were likely due to three additional hours of light exposure. Likewise, mean 24-h plasma melatonin concentrations were decreased after 6 nights of 4-h versus 12-h bedtimes.…”

“…NAS produced by the N ‐acetylation of serotonin displays marked diurnal rhythms 9 both in rats 10 and humans 11 with higher levels at night in all species. NAS can be deacetylated to form serotonin by rice histone deacetylase 10 and its Arabidopsis ortholog, histone deacetylase 14 12 . It is unknown whether NAS can be deacetylated to form serotonin in mammals.…”
